Summary: The study revealed that overexpression of HO-1 in microglial cells in aged mice leads to neurotoxic iron accumulation, oxidative stress, and cognitive decline. Inhibiting HO-1 or using an iron chelator can prevent these issues, providing new insights for treating age-related neurodegenerative diseases.

Summary: Neuropathic pain causes severe pain and comorbidities such as cognitive impairment, which are not effectively treated with current analgesics. This study demonstrates the role of interleukin-6 (IL-6) in neuropathic pain and cognitive impairment. Blocking the IL-6 receptor improves cognitive impairment, while inhibiting IL-6 activity reduces allodynia.

Summary: A large number of people with HIV experience chronic widespread pain. This study found that high heme or low HO-1 levels caused mast cell activation and release of pain mediators in both HIV patients and animal models. CO donation inhibited mast cell degranulation and reduced pain sensitivity in the animal models.

Summary: This study analyzed the analgesic effects of three new HO-1 inducers and dimethyl fumarate on neuropathic pain and investigated their impact on anxiety, depression, and antioxidant enzymes. The results showed that these compounds effectively alleviated pain and emotional disorders by modulating antioxidant and inflammatory pathways.

Summary: This review summarizes the existing preclinical evidence on how the CNS compartment and sex impact microglia functions in health, emphasizing recent advances in transcriptomics analyses for characterizing disease-associated microglial states. The specific scenario of peripheral nerve or tissue injury inducing expression of a specific subset of genes in microglia in the dorsal horn of the spinal cord is discussed, along with the possibility of a unique microglia transcriptional profile associated with chronic pain conditions. The evidence also suggests microglial activation in pain-related areas of the brain can be observed in models of neuropathic pain, highlighting the potential for future studies to further explore neuroimmune interactions in chronic pain using novel approaches like scRNA-seq and CYTOF.

Summary: By using RNA sequencing and experimental validation, this study identified that TPSD1 gene induces the activation of the NF-kappa B signaling pathway, leading to the activation and M1 polarization of microglia, secretion of pro-inflammatory factors, and ultimately resulting in neuropathic pain. Targeting TPSD1 could improve the management of neuropathic pain.

Summary: The study indicates that spinal HO-1 plays a crucial role in the analgesic effects of calcium channel alpha(2)delta ligands by attenuating glial activation and promoting endogenous opioid release. Additionally, peripheral HO-1 inhibition does not affect the analgesic efficacy of pregabalin and gabapentin.

Summary: The cGAS-STING pathway may be a potential therapeutic target for treating neuropathic pain by suppressing microglial M1 polarization. Activation of the cGAS-STING pathway occurs mainly in neurons and microglia, and inhibition of the pathway can suppress microglial M1 polarization and improve mechanical allodynia.

Summary: Neuropathic pain is a chronic condition that affects 7-10% of the general population and is often undertreated. Cannabinoids show promise as a potential treatment for neuropathic pain, but CB1 agonists have serious side effects. Recent studies have found that CB2 agonists can effectively relieve neuropathic pain without causing psychostimulant effects. A novel selective CB2 agonist, COR167, has been shown to attenuate mechanical allodynia and thermal hyperalgesia without tolerance induction, making it a promising therapeutic option.

Summary: NP is a common accompanying symptom of many diseases, seriously affecting the quality of life of patients. Neuron-microglia interaction has been found by experts to play a vital role in the occurrence and development of NP. Advanced studies and interventions using pharmaceuticals and traditional herbal medicines extracts have been attempted to address NP due to its unsatisfactory clinical treatments.
