Summary: Melanoma, a highly heterogeneous tumor, can dynamically shift between different transcriptional states or phenotypes in a process known as phenotype switching. This switch involves changes in cell cycle states, invasiveness, signaling pathways, and immune cell composition. Melanoma cell plasticity is driven by gene expression changes in immune cells, cancer-associated fibroblasts, and alterations in the extracellular matrix, which contribute to metastasis and therapy resistance. This review highlights recent findings on melanoma phenotype switching, immunotherapy resistance, and the role of neural crest-like states in melanoma.

Summary: Combined therapy with anti-BRAF plus anti-MEK is the first-line treatment for metastatic melanomas. However, tumor cell resistance is a major issue. This study shows that DDR2 inhibition can reduce AXL expression and stress fiber formation in resistant melanoma cells. Furthermore, DDR2 inhibition can control cell and tumor proliferation through the MAP kinase pathway.

Summary: By blocking the MNK1/2-eIF4E axis, the study demonstrated inhibition of melanoma phenotype switching and increased sensitivity to anti-PD-1 immunotherapy in melanoma mouse models. Phospho-eIF4E-deficient melanomas expressed high levels of melanocytic antigens, and genetically ablating phospho-eIF4E reprogrammed the immunosuppressive microenvironment. Dual blockade of MNK1/2-eIF4E and PD-1/PD-L1 immune checkpoint showed efficacy in multiple melanoma models and led to an increase in intratumoral stem-like TCF1(+)PD-1(+)CD8(+) T cells.
