In vitro and in vivo anti-diabetic activity of Swertia kouitchensis extract

Ethnopharmacological relevance: Swertia kouitchensis has long been used as a folk medicine to treat hepatitis and diabetes in central-western China. Therefore, this study was aimed to evaluate the antidiabetic activity of the plant ethanol extract. Materials and methods: Firstly, the extract was tested for its inhibitory activity on alpha-amylase and alpha-glucosidase in vitro. Following that, insulin secretion test in NIT-1cell was performed. Then, oral sucrose or starch tolerance test of the extract were carried out in normal mice. After that, acute effect of the extract was executed in normal and streptozotocin-induced (60 mg/kg) diabetic mice. Eventually, long term effect of the extract was performed in diabetic mice for 4 weeks. Oral glucose tolerance test and biochemical parameters were estimated at the end of the study. Results: Swertia kouitchensis extract could remarkably inhibit the activity of alpha-amylase and alpha-glucosidase and stimulate insulin secretion in vitro. And also the extract displayed anti-hyperglycemic activity, improved antioxidant capacity, ameliorated the hyperlipidemia and carbohydrate metabolism in diabetic mice. Conclusions: Swertia kouitchensis exhibits considerable anti-diabetic activity and metabolic alterations in diabetic mice. These results provide a rationale for the use of Swertia kouitchensis to treat diabetes mellitus. (c) 2013 Elsevier Ireland Ltd. All rights reserved.

Summary: The study demonstrated that Xiaokeyinshui extract combination (XEC) has hypoglycemic effects against Type 2 diabetic mellitus (T2DM) by increasing insulin secretion, improving HDL-C levels, reducing biochemical indicators, and repairing islet structure. Analysis of plasma metabolic profiles suggested that XEC may regulate carbohydrate metabolism, lipid metabolism, and amino acid metabolism in the treatment of T2DM.

Sesquiterpenoids from Ixeris sonchifolia and their neuroprotective activities

Summary: The phytochemical investigation of the methanol extract of Ixeris sonchifolia resulted in the isolation and identification of nine analogs, including a new guaiane-type sesquiterpenoid, ixerinoid A. Compound 1's structure was determined through extensive analysis and quantum chemical calculations. Compounds 3, 5, 6, 8, and 9 demonstrated significant protective effects in a neuroprotective activity test.

Jatrophane Diterpenoids with Kv1.3 Ion Channel Inhibitory Effects from Euphorbia helioscopia

Summary: In this study, a total of 17 new jatrophane diterpenoids, named heliojatrone D (1) and helioscopids A-P (2-17), were isolated and identified from the whole plant of Euphorbia helioscopia. Additionally, 11 known analogues (18-28) were also found. The inhibitory effect of these compounds against Kv1.3 ion channels was evaluated, and two compounds (2 and 15) showed the most potent activity with IC50 values of 0.9 μM.

Brujavanoids A-U, structurally diverse apotirucallane-type triterpenoids from Brucea javanica and their anti-inflammatory effects

Summary: A total of 27 triterpenoids, including 21 previously undescribed compounds, were isolated and identified from the methanol extract of the inflorescences, twigs, and leaves of Brucea javanica. Among them, brujavanoid A is the first apotirucallane-type triterpenoid with a novel 19(10 & RARR; 9)abeo motif, and brujavanoids B and C are the first apotirucallane-type triterpenoids with a rarely occurring 14-hydroxy-15,16-epoxy fragment. The most active compound, brujavanoid E, exhibits significant anti-inflammatory effects.

Anti-inflammatory lanostane triterpenoids with rearranged spirobi[indene] scaffold and their biogenetically related analogues from Euphorbia maculata

Summary: Phytochemical investigation of Euphorbia maculata Linn. yielded 16 lanostane-related triterpenoids, including 11 previously undescribed compounds. The structures of these compounds were determined through spectroscopic data interpretation, quantum chemical calculation, and X-ray crystallography. The isolated compounds showed inhibitory effects on NO release in LPS-activated RAW264.7 cells, with spiromaculatol C (3) exhibiting the most potent activity by reducing NF-kappa B p65 nuclear translocation.

This study describes the development of a practical approach for identifying snake gallbladder crude drugs, which can effectively distinguish samples from different species.

Ameliorative effect of scopolamine-induced cognitive dysfunction by Fufangmuniziqi formula: The roles of alkaloids, saponins, and flavonoids

Summary: This study investigated the effects of Fufangmuniziqi formula (FFMN) on Alzheimer's disease (AD) and the role of alkaloids, flavonoids, and saponins in the process. The results showed that FFMN improved cognitive and memory impairment in the AD mouse model. Alkaloids enhanced the cholinergic system, while flavonoids and saponins attenuated neuroinflammation and oxidative stress. These findings suggest that the combination of alkaloid, flavonoid, and saponin fractions derived from FFMN is a promising anti-AD agent.

This study analyzed the phytochemical profile of the aqueous extract of M. longifolia and evaluated its acute and subacute toxicity in mice. The results showed that M. longifolia extract is rich in phenolic compounds and has no observed toxicity up to a certain dosage limit.

A study integrated metabolomics and network pharmacology to investigate the effects of Shicao in alleviating acute liver injury

Summary: This study aimed to explore the hepatoprotective effects of Shicao on CCl4-induced acute liver injury. A rat model of acute liver injury was established and liver function indices were assessed. Untargeted metabolomics was conducted to identify differential metabolites. Network pharmacology was used to construct a network of metabolite-reaction-enzyme-gene constituents, and key components and hub genes were screened. Shicao showed hepatoprotective effects against acute liver injury in rats.
