Summary: This study reveals the role of caspase-8 in regulating IL-1 beta activation in macrophages and its involvement in pulmonary arterial hypertension (PAH). Caspase-8 promotes inflammatory cell infiltration and pulmonary arterial smooth muscle cell proliferation via NLRP3/IL-1 beta activation, contributing to the pathogenesis of PAH.

Summary: Monocytes and monocyte-derived macrophages infiltrate and promote the growth of glioblastoma. PDGFB-driven GBM cells induce the expression of IL-1 beta in MDM, which activates the NF-kappa B pathway and leads to the induction of monocyte chemoattractant proteins. This feedforward circuit between IL-1 beta/IL-1R1 in tumors and MDM drives PDGFB-driven GBM progression.

Summary: This study characterized the effects of miR-21 delivered by exosomes from MSCs on macrophage polarization in a mouse sepsis model. Beta MSC-derived exosomes induced M2-like polarization of macrophages more effectively, improving survival rates in septic mice. Exosomal miR-21 was identified as a key mediator of the immunomodulatory properties of MSCs induced by IL-1 beta pretreatment, providing a potential therapeutic basis for sepsis.

Summary: The study revealed that tannic acid (TA) inhibits IL-1 beta-induced inflammation and matrix degradation in human OA chondrocytes by blocking IL-1 beta-IL-1R1 interaction. In addition, TA showed protective effects in a rat OA model by reducing pain, cartilage degradation, and IL-1 beta-mediated inflammation.

Summary: Treatment of cancer cachexia is still an unmet need, with recent trials showing a potential role for targeting IL-1 beta using monoclonal antibody canakinumab in lung cancer. The neuroinflammation mediated by IL-1 beta plays a critical role in cachexia development, leading to muscle proteolysis and adipose lipolysis, but the precise mechanism of targeting IL-1 beta in treating cachexia remains unclear.

Summary: In this study, it was found that interleukin 1 beta (IL-1 beta) induces the expression of interleukin 8 (IL-8) in canine cardiac fibroblasts through the activation of nuclear factor (NF)-kappa B p65.

Summary: This study reveals the role of BRG1 in embryonic lymphatic development and highlights the epigenetically regulated crosstalk between macrophages, blood vessels, and lymphatics. The findings suggest that IL-1 beta, produced by omental macrophages, destabilizes adherens junctions in omental blood and lymphatic vessels, leading to erythrocyte extravasation and uptake by lymphatics.

Summary: The bidirectional interaction between adipocytes and adipose tissue immune cells is crucial for regulating adipose function, inflammation, and insulin responsiveness. Obesity reduces cAMP-driven IL-6 secretion from adipocytes, but restoring their catecholamine sensitivity can stimulate IL-6 secretion and activate macrophages to promote an anti-inflammatory gene expression pattern.

Summary: The study evaluated the role of skin bacteria in wound healing and hair follicle regeneration, finding a correlation between bacterial counts and WIHN levels. Reduction of skin microbiota decreased WIHN, while IL-1 beta and IL-1R-MyD88 signaling played crucial roles in bacteria-promoted regeneration.

Summary: By studying zebrafish kidney macrophages during A. hydrophila infection, we found that the canonical Wnt signaling pathway is activated, leading to mitochondrial fission, cell apoptosis, and bacterial clearance. This study reveals the host-centric role of the canonical Wnt signaling pathway in A. hydrophila pathogenesis.

Summary: Sepsis is a life-threatening syndrome, and its associated mortality is increased when cardiac dysfunction and damage (septic cardiomyopathy [SCM]) occur. The NLRP3 inflammasome plays a critical role in the pathophysiology of SCM, with the activation of IL-1 beta and IL-18 contributing to the development of SCM.
