Quantitative Detection of Disseminated Melanoma Cells by Trp-1 Transcript Analysis Reveals Stochastic Distribution of Pulmonary Metastases

Lenka Kyjacova, Rafael Saup, Melanie Rothley, Anja Schmaus, Tabea Wagner, Anja Bosserhoff, Boyan K. Garvalov, Wilko Thiele, Jonathan P. Sleeman

Summary: A better understanding of melanoma metastasis is crucial for developing novel therapies. The use of qRT-PCR for sensitive quantification of Trp-1 in murine lung metastasis allows detection even in low metastatic burdens, enabling quantification and analysis in various melanoma models.

Sulfated hyaluronic acid inhibits the hyaluronidase CEMIP and regulates the HA metabolism, proliferation and differentiation of fibroblasts

Anja Schmaus, Melanie Rothley, Caroline Schreiber, Stephanie Moeller, Sven Rosswag, Sandra Franz, Boyan K. Garvalov, Wilko Thiele, Sofia Spataro, Carsten Herskind, Marco Prunotto, Ulf Anderegg, Matthias Schnabelrauch, Jonathan Sleeman

Summary: This study reveals the importance of CEMIP, a hyaluronidase enzyme, in the metabolism, proliferation, and differentiation of fibroblasts. Inhibiting CEMIP with a compound called sHA3.7 increases cellular levels of hyaluronan and promotes fibroblast differentiation into myofibroblasts.

Increased Circulating Osteopontin Levels Promote Primary Tumour Growth, but Do Not Induce Metastasis in Melanoma

Rafael Saup, Nidhi Nair, Jingyi Shen, Anja Schmaus, Wilko Thiele, Boyan K. Garvalov, Jonathan P. Sleeman

Summary: The role of circulating osteopontin (cOPN) in melanoma progression was studied. It was found that increased cOPN promotes primary tumor growth but does not significantly affect spontaneous metastasis of melanoma cells to lymph nodes or lungs.
