Summary: Limb and CNS expressed 1 like (LIX1L) is over-expressed in various tumors, especially hepatocellular carcinoma (HCC). Overexpression of LIX1L enhances proliferation, migration, and invasion of HCC cells, while reducing glucose consumption and lactate production. Through upregulation of miR-21-3p and suppression of FBP1, LIX1L promotes HCC growth and metastasis. Inhibition of miR-21-3p could suppress tumor growth in an orthotopic tumor model.

Disrupting IL23R enhances Treg responsiveness to IL12, resulting in increased production of γ-interferon and recruitment of CD8 T cells that inhibit tumor growth. Combined targeting of destabilization pathways in Treg can lead to robust and synergistic antitumor responses.

Summary: Understanding the mechanisms of how T cells become dysfunctional in a tumor microenvironment is crucial for cancer immunotherapy. This study found that CD36 expression in tumor-infiltrating CD8(+) T cells, induced by TME cholesterol, is associated with tumor progression and poor survival, and that genetic ablation of Cd36 in these T cells leads to enhanced tumor eradication. Targeting CD36 or inhibiting ferroptosis could restore T cell function and enhance antitumor efficacy, especially in combination with anti-PD-1 antibodies.

Summary: Liver fibrosis is an inevitable stage in the development of chronic liver disease to cirrhosis. Interventional treatment and controlling the disease at this stage can reduce the incidence of liver cirrhosis. The expression of VDR in liver tissues of patients with liver fibrosis is negatively correlated with alpha-SMA, Col-1, and liver fibrosis stages. Calcipotriol, a VDR agonist, effectively reduces the degree of liver fibrosis by inhibiting the deposition of ECM and activation of HSCs.
