Summary: The study aimed to investigate the effect of miR-369-3p on the expression and activation of NLRP3 inflammasomes via BRCC3 regulation. The findings showed that increased miR-369-3p significantly reduced the expression of BRCC3, NLRP3, and ASC adaptor protein, resulting in decreased Caspase-1 activity and reduced cleavage of pro-IL-1β and pro-IL-18.

Summary: This study investigated the mechanisms of NLRP3-mediated macrophage pyroptosis and identified the role of miR-223-3p in regulating NLRP3 expression. It was found that miR-223-3p can decrease the expression of NLRP3 and proinflammatory factors, resulting in reduced pyroptosis in irradiated murine macrophages. In vivo experiments further demonstrated the effectiveness of miR-223-3p supplementation in alleviating acute radiation-induced lung injury.

Summary: This study shows that Atranorin, a secondary metabolite of lichen family, effectively inhibits NLRP3 inflammasome activation in macrophages and dendritic cells. It binds to ASC protein directly and restricts ASC oligomerization, leading to the suppression of cytokine secretion and cell pyroptosis. Atranorin exhibits therapeutic effects in various NLRP3 inflammasome-driven diseases.

Summary: By screening against NLRP3-dependent pyroptosis, a novel compound J114 was discovered, which displayed significant inhibitory activity against human cell pyroptosis and showed distinct differences from existing pyroptosis inhibitors. Further studies revealed that J114 inhibited inflammasome activation by disrupting the interaction between NLRP3 or AIM2 and the adaptor protein ASC.

Summary: Diabetic nephropathy (DN) is a microvascular complication of diabetes. Pyroptosis of renal tubular epithelial cells plays a critical role in DN. Mesenchymal stem cells (MSCs) have immunomodulatory properties and can ameliorate kidney damage and reduce inflammation in DN rats. miR-342-3p, which is downregulated in DN kidneys, inhibits pyroptosis by targeting the NLRP3/Caspase1 pathway.

Summary: The study reveals that T. marneffei yeasts induce IL-1 beta production, with NLRP3 inflammasome playing a crucial role in host immune response. Dectin-1/Syk signaling pathway mediates IL-1 beta production, and NLRP3-ASC-caspase-1 inflammasome facilitates the processing of pro-IL-1 beta into IL-1 beta. Mice deficient in NLRP3 or caspase-1 show higher mortality and fungal load after T. marneffei infection, indicating the important role of NLRP3 inflammasome in host defense.

Summary: The activation of the inflammasome plays a critical role in the pathogenesis of systemic lupus erythematosus (SLE), and mesenchymal stem cells (MSC) have been shown to alleviate SLE by suppressing inflammasome activation. This study found that the NLRP3 inflammasome was activated in macrophages from SLE patients and mice, and its activation correlated with disease activity. After MSC transplantation, the severity of SLE was reduced, and NLRP3 inflammasome activation was inhibited. These findings suggest that MSC suppress inflammasome activation and provide a potential therapeutic target for SLE.
