Summary: The aim of this study was to investigate the effect of apigenin on pyroptosis and explore its potential against inflammatory diseases. The results showed that apigenin exhibited a preventive effect on macrophage pyroptosis by reducing oxidative stress and inhibiting the NF-κB pathway. These findings suggest apigenin to be a promising therapeutic agent for inflammatory diseases.

Summary: This study found that hyperhomocysteinemia can lead to larger atherosclerotic plaques and increased secretion of inflammatory cytokines, which can be attenuated by reducing Caspase-1 levels in mice. In vitro experiments demonstrated that homocysteine can activate the NLRP3 inflammasome and induce macrophage pyroptosis, which is inhibited by Caspase-1 inhibition. Furthermore, homocysteine promotes endoplasmic reticulum stress, endoplasmic reticulum-mitochondria coupling, and disturbs calcium homeostasis, contributing to macrophage pyroptosis.

Summary: Pseudorabies virus (PRV) preferentially invades neural tissue and organs, causing multisystemic lesions. In this study, it was shown that PRV infection triggers GSDMD-mediated pyroptosis in porcine alveolar macrophage cells through the activation of caspase-1 and the cleaving of GSDMD. NLRP3 and IFI16 inflammasomes were also activated, contributing to pyroptosis during PRV infection. These findings enhance our understanding of the inflammatory response and cell death pathways caused by PRV.
