The Golgi apparatus is a dynamic organelle, which regulates the vesicular trafficking. While cellular trafficking requires active changes of the Golgi membranes, these are not accompanied by changes in the general Golgi's structure. However, cellular processes such as mitosis, apoptosis and migration require fragmentation of the Golgi complex. Currently, these changes are most commonly studied by basic immunofluorescence and quantified by manual and subjective classification of the Golgi structure in 100-500 stained cells. Several other high-throughput methods exist as well, but those are either complicated or do not provide enough morphological information. Therefore, a simple and informative high content methodology should be beneficial for the study of Golgi architecture. Here we describe the use of high-throughput imaging flow cytometry for quantification of Golgi fragmentation, which provides a simple way to analyze the changes in an automated, quantitative and non-biased manner. Furthermore, it provides a rapid and accurate way to analyze more than 50,000 cells per sample. Our results demonstrate that this method is robust and statistically powerful, thus, providing a much-needed analytical tool for future studies on Golgi dynamics, and can be adapted to other experimental systems.

Summary: Combining single-cell RNA sequencing with spatial information allows for the reconstruction of spatially-resolved tissue cell atlases, which is challenging for rare cell types. The ClumpSeq approach presented in this study enables the establishment of spatial atlases for all epithelial cell types in the small intestine, revealing immune-modulatory gene expression in villus tip goblet and tuft cells as well as heterogeneous migration patterns of enteroendocrine cells.

Visual barcodes for clonal-multiplexing of live microscopy-based assays

Summary: In this study, the authors developed a fluorescence imaging-based visual barcode for livecell clonal-multiplexing, which allows the identification of signalling pathways clusters in response to different chemotherapy compounds. They also demonstrated the use of visual barcodes to generate 'Signalome' cell-lines, enabling the simultaneous monitoring of the activity in 12 branches of signaling at clonal resolution.

Summary: HNRNPU loss of function leads to rapid cell death of both postmitotic neurons and neural progenitors, with a higher sensitivity of the latter. The expression and alternative splicing of multiple genes involved in cell survival, cell motility, and synapse formation are affected following Hnrnpu's conditional truncation. Pharmaceutical and genetic agents have been identified to partially reverse the loss of cortical structures in Hnrnpu mutated embryonic brains.

Summary: The accumulation of fatty bone marrow and somatic mutations in hematopoietic stem cells (HSCs) increases with aging. In preclinical mouse models, it is found that fatty bone marrow can enhance the self-renewal of HSCs and activate inflammatory signaling. The inflammatory cytokines, such as IL-6, in fatty bone marrow promote DNMT3a-driven clonal hematopoiesis, which can be inhibited by blocking the IL-6 pathway.
