Summary: Chronic rhinosinusitis with nasal polyps (CRSwNP) is caused by prolonged inflammation of the paranasal sinus mucosa. In this study, TIM-4 was found to be increased in the sinonasal mucosa of CRSwNP patients, especially in macrophages. TIM-4 was positively correlated with clinical scores and inhibition of TIM-4 reduced polyp formation and EMT.

Summary: The role of Foxp3 in peripheral pTreg cells and the mechanisms supporting their differentiation remain poorly understood. This study used genetic tracing to identify microbiota-induced pTreg cells and found that many of their distinguishing features were Foxp3 independent. While Foxp3 was critical for the suppression of certain diseases, pTreg cells could suppress colonic effector T cell expansion in a Foxp3-independent manner. Thus, Foxp3 and the tolerogenic signals preceding and promoting its expression confer distinct facets of pTreg functionality.

Summary: The spatial organization of immune cells within the tumor microenvironment (TME) plays a crucial role in anti-tumor immunity, tumor progression, and therapeutic response. Tim-3 is an immune checkpoint with potential as an immunotherapy target. This study examined the spatial distribution of Tim-3/Tim-3L in the TME and found that CD4 T cells were more accumulated in the tumor region and had a closer association with patient survival. CD4 T cells surrounding Tim-3L(+) cells expressed higher levels of Tim-3. Depletion of CD4 T cells abrogated the inhibition of tumor growth and metastasis mediated by Tim-3 blockade. These findings highlight the importance of CD4 T cells in Tim-3/Tim-3L-mediated immunosuppression and provide insights for Tim-3 targeted cancer immunotherapy.

Summary: This study found that acute and chronic HIV infection are associated with an increase in total, effector memory, and terminally differentiated FoxP3(+) CD8 T-cells. Early antiretroviral therapy (ART) only normalized the frequencies of total FoxP3(+) CD8 T-cells. The study also observed increased immune activation, senescence, and PD-1 expression in FoxP3(+) CD8 T-cells during acute and chronic infection, which were not normalized by early ART.
