Summary: In effector T cells, the pore subunit of CRAC channels, ORAI1, is limited in plasma membrane localization with a significant fraction trapped in intracellular vesicles. NKD2, an essential component of ORAI1(+) vesicles activated by signaling pathways downstream of T cell receptors, orchestrates trafficking and insertion of ORAI1(+) vesicles to the plasma membrane. T cell receptor (TCR)-stimulation-dependent insertion of ORAI1 into the plasma membrane is essential for sustained calcium signaling and cytokine production in T cells.

Summary: Non-genetic photostimulation is a growing multidisciplinary field that aims to induce light-sensitivity in living systems using exogeneous phototransducers. In this study, an intramembrane photoswitch (Ziapin2) was proposed for optical pacing of hiPSC-CMs. The effects of light-mediated stimulation on cell properties were investigated, including changes in membrane capacitance, membrane potential (V-m), intracellular Ca2+ dynamics, and cell contractility. The results demonstrate that Ziapin2 can modulate electrical activity and contractility in hiPSC-CMs, offering potential for future cardiac physiology development.

Summary: This article describes a potentially unique cytokine designed to separate T cell surface binding and signaling, thus improving the side effects and cell exhaustion associated with cytokine therapy. The fusion protein cytokine showed precise activation of cytotoxic T cells, improved their survival and memory cell generation, and decreased exhaustion. This functional improvement was achieved through altered signal transduction pathways, which increased CD8(+) T cell response to low-affinity antigens, activated transcription factors, and promoted mitochondria biogenesis.

Summary: The interplay between T- and B-cell compartments during naive, effector and memory T cell maturation is critical for a balanced immune response. Primary B-cell immunodeficiency arising from X-linked agammaglobulinemia (XLA) offers a model to explore B cell impact on T cell subsets, starting from the thymic selection. The findings suggest active B cell involvement in CD4 T cell subsets maturation, including B cell-dependent expansion of the naive Treg TCR repertoire that enables better control of self-reactive T cells.

Summary: Nanotechnology-enabled neuromodulation is a promising minimally-invasive tool that can be used in neuroscience and engineering for both fundamental studies and clinical applications. This study reveals the transformation of neuromodulation from heterogeneous to homogeneous in a progressively maturing neural network, and the selective nano-neuro interaction between nanoparticles and neurons based on their surface charge. The density of nanoparticles bound to neurons in a developing neural network leads to heterogeneous optical neuromodulation, which simultaneously excites and inhibits neural network activity.
