Summary: In this paper, a microfluidic device is developed to study the sprouting angiogenesis of endothelial cells under oxygen gradients and co-culture of fibroblast cells. The device integrates a hydrogel matrix for cell culture and generates stable oxygen gradients using a chemical reaction method. The results show that the oxygen gradient and co-culture of fibroblast cells promote sprouting of endothelial cells by altering cytokines and hydrogel properties. This device provides a powerful in vitro model to investigate sprouting angiogenesis under in vivo-like microenvironments.

Summary: This study used label-free surface-enhanced Raman spectroscopy (SERS) method and micro-plasma treatment to detect membrane proteins in exosomes secreted by melanoma cells. The results showed that micro-plasma treatment increased extracellular ROS/RNS levels and slightly reduced cell proliferation. The particle size of secreted exosomes may change under different treatment times and co-culture conditions. In addition, the SERS method can identify characteristic peaks related to exosome markers CD9, CD63, and CD81.

Summary: This article describes a hemicyanine hybrid probe containing 7-(diethylamino)coumarin (ACou) donor and cationic indolenium (Ind) acceptor, which is used for detecting nucleophilic analytes such as cyanide and reactive sulfur species (RSS). The study reveals that thiolates react with ACou-Ind through conjugate addition to form C4-SR adducts, and ACou-Ind serves as a turn-off probe by losing red ICT fluorescence upon RSS addition. Moreover, the article discusses that the blue coumarin emission of thiolate adducts does not enhance in the presence of reactive oxygen species (ROS), but undergoes oxidative deconjugation in the presence of ROS H2O2. The relevance of the chemosensing mechanism for ACou-Ind is assessed in live zebrafish, and the implications for the interplay between RSS and ROS are discussed.

Summary: In this study, the activation of SPC using micro amounts of Cu(II) for AO7 degradation was explored. The results showed that nearly 97.5% of AO7 could be removed within 15 minutes using 20μM Cu(II) and 0.4 mM SPC. The Cu(II)/SPC system could work under near-neutral or alkaline solutions, making it more advantageous than conventional Fenton oxidation.

Summary: The study aimed to determine whether the antioxidant amino acid N-acetyl cysteine (NAC) reduces the toxicity of dental restorative materials. The results showed that impregnating certain materials with NAC can mitigate their toxicity and improve the attachment, growth, and function of human oral fibroblasts on the materials. These improvements are primarily achieved by reducing the production of reactive oxygen species and enhancing the cellular antioxidant defense system.

Summary: Although the treatment efficacy for hematological malignancies has improved, relapse or drug resistance remains a challenge. Microenvironment disorder plays a significant role in the pathogenesis of these malignancies. Immunogenic cell death (ICD) is a unique mechanism that triggers an adaptive immune response through danger signals or damage-associated molecular patterns, making it a potential immunotherapeutic approach for hematological malignancies. This review provides an overview of ICD induction in hematological malignancies and current research on combining ICD inducers with other treatment strategies.

Summary: This study revealed that fibroblast-like synoviocytes (FLS) from rheumatoid arthritis (RA) patients had increased levels of reactive oxygen species (ROS) and impaired redox balance compared to FLS from control patients. Liver kinase B1 (LKB1) was found to play a crucial role in scavenging ROS and cellular metabolism in various cancers. Through siRNA-induced LKB1 deficiency, it was found that LKB1 regulated the AMPK-mediated SLC7A11-NOX4-ROS pathway to control FLS migration and inflammation in RA. This indicates that LKB1 is a key regulator of redox homeostasis in RA FLS.

Summary: The study evaluated the chemical composition and cytotoxic effects of acai seed extract on breast cancer cell line MCF-7. It was found that acai seed extract showed cytotoxic effects against MCF-7 cells, induced morphologic changes through autophagy, and increased ROS production. In addition to its antioxidant activity, flavonoids in acai also generated NO, contributing to its cytotoxic effects and autophagy induction.

Summary: Different isolation methods of tendon cells have an impact on cell behavior, fate, and lineage composition. Cells isolated by explant exhibit reduced proliferative capacity, decreased expression of tendon marker genes, and increased expression of genes associated with fibroblast activation compared to digested cells. Explanted cells also have an increased propensity to differentiate to myofibroblasts compared to digested cells.
