Summary: Depression is a prevalent psychiatric disorder associated with chronic stress, monoaminergic dysfunction, impaired neurogenesis, and decreased BDNF-CREB signaling. The study suggests that F3/Contactin, a cell-adhesion molecule, may be implicated in depression and could serve as a novel antidepressant target. Over-expression of hippocampal F3/Contactin prevents depressive-like behaviors and dysfunction in neurogenesis and signaling, while administration of vortioxetine, an antidepressant, ameliorates the inhibitory effects on F3/Contactin expression.

Summary: Duman's research has shown that long-term treatment with antidepressant drugs increases neurogenesis in the subgranular zone of the adult hippocampus, providing critical insights into the neurotrophic hypothesis of depression and synaptic plasticity. It has been demonstrated that neurogenesis may be necessary for the clinical effects of antidepressant drugs, and his lab continues to investigate neurotrophic factors and synaptic plasticity. Preclinical studies have also shown that newer antidepressant compounds impact neurogenesis and synaptic plasticity.

Summary: Depression, one of the most common psychiatric diseases in the 21st century, is not fully understood in terms of its pathogenesis. This study suggests that the brain-derived neurotrophic factor (BDNF)-cAMP response element-binding protein (CREB) signaling pathway is attractive for treating depression. In particular, the research finds that hippocampal MSK1 plays an important role in the pathogenesis of depression and could be a potential target for antidepressant treatment.

Summary: The study showed that neonatal exposure to Sev anesthesia induced cognitive impairment in adult rats, reducing the number of neurons and the concentration of BDNF in the hippocampus. Furthermore, Sev treatment decreased the expression of PKA, p-CREB, BDNF, and the p-CREB/CREB ratio, indicating the importance of PKA-CREB-BDNF signaling in cognitive decline caused by Sev exposure.

Summary: Estrogen deprivation during menopause leads to changes in brain neuronal circuit and neurobehavioral symptoms later in life. Hormone replacement therapy can partially alleviate these abnormalities but is associated with various adverse effects. This study investigated the effect of lithium therapy on neurobehavioral impairments in a rat model of ovariectomy, which mimics postmenopausal conditions. The results showed that lithium treatment protected against spatial and recognition memory impairments, as well as depression-like behavior in ovariectomized rats. Additionally, lithium treatment increased dendritic length and spine density in certain brain regions, reduced proinflammatory markers, normalized reactive astrogliosis, and modulated key signaling pathways. These findings suggest that lithium may be a potential therapeutic agent for managing postmenopausal neurological symptoms.
