Summary: The study revealed that knockdown of CISD2 accelerated ferroptotic cell death induced by Erastin, characterized by increased lipid peroxidation, GSH exhaustion, and iron accumulation. CISD2 depletion enhanced lysosomal activity and activated ferritinophagy under Erastin exposure. Additionally, CISD2 knockdown inhibited the p62-Keap1-NRF2 pathway, leading to oxidative stress and ferroptosis.

Summary: Anti-cancer therapy by iron chelation using SK4 can inhibit multiple cellular processes and target iron addiction and LAT1 overexpression. The mode of action of SK4 was characterized using proteomics, metabolomics, lipidomics and seahorse real-time analysis, revealing impaired energy metabolism and a potential role of ceramide induction in the phenotypic outcome. These findings highlight the effectiveness of SK4 as a double pronged approach towards treating cancer.

Summary: The resistance of bacteria to antibiotics is a serious problem that limits their effective use. This study focused on the killing of Staphylococcus aureus by ciprofloxacin and the effects of Reactive Oxygen Species (ROS) inactivator thiourea and iron chelator DIBI on bacterial survival. The results showed that these agents could influence the rate of killing and recovery growth of bacteria upon exposure to ciprofloxacin.

Summary: Acute lung injury (ALI) has been a challenging issue in healthcare systems for a long time, and effective prevention and treatment strategies are still lacking. Iron dysregulation is a common characteristic in many subtypes of ALI, playing a dual role in immune response and host cell damage. Iron chelation offers a novel therapeutic approach for alleviating lung injury and improving the survival of ALI patients. This article provides an overview of iron homeostasis, the role of iron in ALI development, and potential therapeutic targets.

Summary: The study proposes the use of a nanocomplex consisting of methylene blue and citrate-coated maghemite nanoparticles (MAGCIT-MB) for the treatment of breast and ovarian cancer. Results from in vitro studies show significant reduction in cell viability of cancer cell lines treated with MAGCIT-MB, without affecting non-tumor cells. This suggests the potential of MAGCIT-MB as a targeted therapy for breast and ovarian cancer.

Summary: This study investigated the mechanism of action of gallium combination therapy in Pseudomonas aeruginosa and its analysis in an in vivo model. The results showed that gallium combination therapy could synergistically kill P. aeruginosa by increasing the organism's susceptibility to oxidative stress. It also demonstrated effective treatment of infection in a murine pulmonary infection model.

Summary: The study identified that the dithiocarbamate derivative PdtaA possesses anti-tumor properties by inducing ferritinophagy-related ROS production, leading to apoptosis, cell cycle arrest, and ferroptosis. This induction of ferroptosis, involving downregulation of GPx4 and xCT, and association with ROS production, represents a novel mechanism of action for a dithiocarbamate derivative.

Summary: The strength of ferritinophagic flux plays a crucial role in regulating EMT and ferroptosis processes, affecting ROS production, modulation of related proteins, and induction of ferroptosis. This study also found that NDRG1 activation is involved in ferroptosis induction, highlighting the complex interplay between ferritinophagy, EMT, and ferroptosis.
