The history of in vitro maturation (IVM) of mammalian oocytes, especially of human oocytes, holds a special place for Robert Edwards. He was the first to comprehensively examine and demonstrate maturation of human oocytes in vitro and in so doing he changed the course of medicine by fertilizing them in vitro. In reviewing his contribution, we have examined the state of the field at the time and discuss his pioneering insights into mammalian oocyte biology. We will also discuss how some of the major concepts and challenges identified by Edwards 50 years ago remain among the major challenges facing IVM today.

Dysregulation of bisphosphoglycerate mutase during in vitro maturation of oocytes

Summary: Hemoglobin and 2,3-BPG may have a role in the maturation of cumulus-oocyte complexes (COCs). The study found that in vitro maturation (IVM) increases the expression of Bpgm within COCs compared to in vivo. Decreased oxygen concentration and the addition of hemoglobin affected Bpgm expression, but not to the levels observed in vivo.

Effect of oxygen and glucose availability during in vitro maturation of bovine oocytes on development and gene expression

Summary: This study found that under low oxygen conditions (7%), oocyte developmental competence is negatively impacted when glucose availability is limited. However, under low oxygen with high glucose concentration or adequate availability, developmental rates can be restored to levels seen in 20% oxygen groups. This highlights the importance of considering glucose concentration and physical culture conditions when comparing the effects of different oxygen concentrations during in vitro maturation.

Fabrication on the microscale: a two-photon polymerized device for oocyte microinjection

The effect of discrete wavelengths of visible light on the developing murine embryo

Summary: Our study investigates the impact of different wavelengths of light on the development of embryos. We found that exposure to the yellow wavelength led to a significant reduction in the number of embryos developing to the blastocyst stage. Increased DNA damage was observed in embryos exposed to the blue, green, or red wavelengths. Only the red wavelength showed a significant decrease in total cell number. Additionally, pregnancy rates were significantly reduced and weight at weaning was significantly higher when embryos were exposed to the red and yellow wavelengths respectively. Our results highlight the wavelength-specific and energy-dependent effects of light on embryo development.

Summary: This study introduces a microfluidic chip that is designed and completely 3D-printed using two-photon polymerization (2PP). It consists of ten channels and ten retrievable cell sample seats, which can enhance cell-surface interactions at the nanoscale and support cell culture. This technology provides the precision needed for nanoliter fluidic devices constructed from multiple interlocking parts.

Oocyte in vitro maturation physiological basis and application to clinical practice

Summary: Oocyte in vitro maturation (IVM) is a minimally invasive technique involving the collection and culture of immature oocytes to maturity outside the body. It is now classified as nonexperimental and is primarily indicated for patients with a high antral follicle count and for fertility preservation in cancer patients. Significant advances in IVM culture methods have been made based on animal studies and a simplified patient treatment protocol, leading to its routine practice in specialized centers worldwide.

Pathogenesis of Reproductive and Metabolic PCOS Traits in a Mouse Model

Summary: PCOS is a common disorder with unclear etiology and pathogenesis, managed based on symptoms. A PCOS mouse model showed that irregular cycles, anovulation, and weight gain are early features, while adiposity, impaired glucose tolerance, and dyslipidemia are later characteristics. These findings support early diagnosis with body weight criteria.
