Summary: In this study, a highly efficient microprojectile transformation system was developed for tannin-containing sorghum by using immature embryos of Hongyingzi as explants. After optimization, a transformation frequency of 27% was achieved and molecular analysis confirmed the presence and expression of transgenes in all transgenic plants. Inheritance analysis further demonstrated the inheritance of transgenes into the next generation, demonstrating the successful establishment of an efficient transformation system for tannin-containing sorghum to overcome the restriction imposed by tannins.

Summary: Genetic transformation is a vital strategy for improving plant biomass and resistance to adverse environments, achieved through imparting desirable genetic characteristics. Research on plant genetic transformation technology can lead to functional analysis of plant genes, efficient utilization of desirable traits, and precise breeding. Various genetic transformation technologies have been developed, including direct methods like Agrobacterium-mediated gene delivery and indirect methods such as particle bombardment, electroporation, silicon carbide whiskers, and pollen tubes. The regeneration of transformed plants usually involves de novo organogenesis or somatic embryogenesis pathways. This review provides an overview of various techniques for plant genetic transformation, their application scope, and limiting factors, serving as a reference for plant transgenic breeding.

Summary: An efficient genetic transformation protocol using a protein fusion between wheat growth-regulating factor 4 (GRF4) and its interacting factor (GIF1) was developed for direct gene editing in elite bread wheat cultivars. Immature embryos of different cultivars were successfully transformed using particle bombardment method, resulting in a significant increase in transformation frequency compared to the control vector. The edited traits, including resistance against leaf rust and powdery mildew, were successfully introduced into the elite cultivars using wheat promoters for gene expression. This protocol has great importance in enabling gene editing in elite wheat cultivars and could be applied to other recalcitrant wheat varieties.

Summary: Genome editing tools based on CRISPR/Cas system have the potential to solve agricultural issues and improve food production. The use of genetic engineering in agriculture has allowed for the introduction of specific traits in crops, while CRISPR/Cas system offers a more precise technique for targeted modification of genes in the host plant genome. The delivery of CRISPR reagents has the potential to overcome limitations with traditional transformation methods and produce transgene-free plants. The CRISPR/Cas system is expected to play a key role in future crop breeding.

Summary: Phytohormones such as cytokinins and auxin play major roles in modulating leaf growth in sesame plants, while a putative antagonistic response between jasmonic acid and salicylic acid was observed during leaf development. Additionally, cytokinins were found to modulate growth arrest during water deficit stress, suggesting a metabolic limitation in cytokinin biosynthesis under drought conditions.
