Summary: Cachexia is common in cancer patients and its molecular basis is still unclear. This study investigated the regulatory pathway of tumors on appetite-inhibiting neurons (POMC) in the hypothalamus. Results showed that the tumor-expressed SEMA3D is positively related to POMC expression. Knocking out SEMA3D in mice increased food intake and body weight, and reduced muscle and fat catabolism. SEMA3D enhances POMC neuron activity by activating NRP2 and PlxnD1 expression. This research reveals that SEMA3D may play a vital role in appetite suppression and catabolic metabolism.

Summary: The aim of this study was to examine the effects of semaglutide on neuropeptide signaling in the hypothalamic energy metabolism of diet-induced obese mice. The results showed that semaglutide improved leptin sensitivity and anorexigenic signaling, while decreasing orexigenic signaling. Furthermore, semaglutide also affected biometric data and hypothalamic gene expressions. Therefore, semaglutide can activate anorexigenic signaling by restoring leptin, amylin, and POMC pathways in diet-induced obese mice.

Summary: Obesity and type 2 diabetes are associated with cognitive dysfunction. This study found that specific neurons in the hypothalamus play an important role in the interface of metabolism and cognition. Acute obesogenic diet administration in mice impaired recognition memory due to defective production of the neurosteroid precursor pregnenolone in the hypothalamus. Our data suggest that pregnenolone's effects on cognitive function were mediated via an autocrine mechanism on POMC neurons, influencing hippocampal long-term potentiation. The relevance of central pregnenolone on cognition was also confirmed in metabolically unhealthy patients with obesity.

Summary: The development of hypothalamic pro-opiomelanocortin (POMC) neurons is crucial in obesity development, but the underlying mechanisms are not well understood. Casein kinase 1 alpha (CK1 alpha) is highly expressed in the embryonic mouse hypothalamus and colocalizes with POMC neurons. Deletion of CK1 alpha in POMC neurons leads to weight gain, metabolic defects, and increased food intake. CK1 alpha deletion decreases the number of POMC-expressing cells, but does not affect apoptosis of POMC neurons. CK1 alpha regulates POMC neuron development via NICD-POMC signaling.

Summary: Aging is a systemic process that poses a risk to physiological functions and eventual death. The molecular mechanisms underlying aging and cognitive decline are not fully understood. Recent findings show that Menin plays a role in neuroinflammation and brain development. In aged mice, there is a decrease in Menin signaling in the hypothalamus, which correlates with systemic aging and cognitive deficits. Restoring Menin expression in the ventromedial nucleus of the hypothalamus in aged mice improves lifespan, learning and memory, and aging biomarkers, while inhibiting Menin in middle-aged mice accelerates aging and cognitive decline. Menin epigenetically regulates neuroinflammatory and metabolic pathways, including D-serine metabolism, and the reduction of Menin in aging leads to impaired D-serine release in the VMH-hippocampus neural circuit. Supplementation of D-serine rescues cognitive decline in aged mice. In conclusion, VMH Menin serves as a key regulator of systemic aging and aging-related cognitive decline.

The results showed that ABA led to a significant decrease in body weight and an increase in activity. Moreover, the number of POMC cells decreased in the ABA group, and the number of orexin A-ir positive cells decreased in the LH in both the D and ABA groups.

Summary: In this study, it was found that Compound-K, an active ingredient in ginseng, inhibits lipid droplet and adipogenesis by downregulating the expression of specific genes and proteins involved in adipocyte differentiation. This compound also exhibits antioxidant properties by decreasing reactive oxygen species production and increasing antioxidant enzymes. The findings suggest that Compound-K exerts its anti-adipogenic effects through the activation of the AMPK signaling pathway and inhibition of the ERK/p38 and AKT signaling pathways.
