Summary: The protein HMCES crosslinks to AP sites in ssDNA to prevent DNA breaks. We discovered a non-proteolytic release mechanism for HMCES-DPCs, regulated by DNA context. HMCES-DPCs are stable in ssDNA but released in dsDNA, allowing downstream repair.

Summary: The co-formulant POE-15 tallow amine (POE-15) and the active ingredient glyphosate in herbicides have been found to be cytotoxic, causing cell necrosis and oxidative stress. The study highlights the importance of comprehensive testing of both co-formulants and active ingredients of commercial pesticides to ensure better regulation and protection of public health and the environment.

Summary: Neurodegenerative diseases involve the loss of specific neurons, which is often caused by environmental pollutants like acrolein. Acrolein is highly active and contributes to oxidative stress, polyamine metabolism, and neuronal damage, thereby impacting the progression of neurodegenerative diseases.

Summary: This article describes the development of an automated device for high throughput sample processing in a comet assay, which reliably assesses DNA damage in individual eukaryotic cells. The device performs as well as manual methods but with the advantages of decreased human involvement and assay run time, making it a valuable approach for high throughput DNA damage assessment.

Summary: Acrolein is a highly reactive toxic aldehyde that can be found in diet and environment. It is associated with various pathological conditions and can induce harmful effects at the cellular level. Polyphenols, found in fruits, vegetables, and herbs, have been shown to protect against acrolein's toxicity by acting as scavengers and regulators. This review discusses the exposure and toxicity of acrolein and the role of polyphenols in mitigating its contamination and health hazards.

Summary: Hirudin, a protein with three disulfide bonds, is commonly used as a model for studying the folding of disulfide-rich proteins. This study investigates the impact of diselenide bridges at different positions on the folding kinetics, yield, and heterogeneity of hirudin, demonstrating enhanced folding rate and yield while reducing complexity. The findings suggest that diselenide substitutions could be a useful tool in designing and characterizing disulfide-rich proteins.

Summary: This study investigated the implication of oxidative stress in small-vessel vasculitis, focusing on acrolein immunohistochemistry in affected skin vessels and systemic stress response. It was found that systemic redox homeostasis and iron metabolism are altered in SVV patients, with possible biomarkers identified for evaluating oxidative status, disease activity, and prevalence. Furthermore, a strong correlation between the accumulation of acrolein-protein adducts in the skin and disease progression was revealed.

Summary: Natural pyrethrins induce oxidative stress, DNA damage and liver toxicity in both human HepG2 cells and zebrafish. The observed chronic toxicity may lead to irreversible DNA damage and severe toxic effects on cells. In conclusion, hepatotoxicity of natural pyrethrins poses a risk to human health by causing liver degeneration and steatosis.

Summary: Gel matrices play a fundamental role in electrophoresis analyses of biopolymers in microscale channels. Both capillary gel and microchannel gel electrophoresis systems have made significant advancements and remain vital tools in bioanalytical chemistry and biotherapeutics. This review summarizes the current state of gels in microscale channels, including traditional and nontraditional polymers, and highlights advances in gel matrices, as well as cutting-edge applications in DNA, RNA, protein, and glycan analyses. Emerging techniques for multifunctional assays in capillary and three-dimensional channels are also discussed.

Summary: In this study, it was found that cadmium exposure induced cell death in HepG2 cells via a ROS-mediated PERK-CHOP-related apoptotic signaling pathway, providing a novel insight into the mechanisms of cadmium-induced hepatotoxicity. Inhibitors targeting oxidative stress and ER stress might be potential strategies for preventing or treating this disorder.

Summary: This study assessed the association between acrolein exposure and pulmonary function, as well as the roles of oxidative DNA damage, inflammation, and pulmonary epithelium integrity. The results demonstrated that acrolein exposure was related to decline in pulmonary function, which was influenced by oxidative DNA damage, inflammation, and pulmonary epithelium injury.
