Distinctive Chromosomal Instability Patterns in Oral Verrucous and Squamous Cell Carcinomas Detected by High-Resolution DNA Flow Cytometry

BACKGROUND: Oral verrucous carcinomas (OVCs) are characterized by better prognosis than oral squamous cell carcinomas (OSCCs). Because chromosomal instability (CIN) in solid tumors is indicative of prognosis, this study investigated whether OVCs and OSCCs were characterized by differences in CIN biomarkers. METHODS: Fresh or frozen multiple tissue samples were submitted to high-resolution DNA flow cytometry (hr DNA-FCM). RESULTS: DNA aneuploid sublines were detected in 6 of 9 OVCs (66.7%) and in 20 of 25 OSCCs (80.0%). Multiple DNA aneuploid sublines were observed, respectively, in 2 of 6 (33.3%) DNA aneuploid OVCs and in 14 of 20 (70%) DNA aneuploid OSCCs (P = .163). OVCs were mainly characterized by DNA Index (DI) values in the near-diploid region (DI not equal 1 and DI < 1.4), whereas aneuploid OSCCs carried most frequently multiple aneuploid sublines with high DI values (DI >= 1.4). DNA near-diploid and high aneuploid sublines were, respectively, 87.5% and 12.5% for the OVCs versus 30% and 70% for the OSCCs (P = .004). CONCLUSIONS: Present data suggest that OVCs are characterized by a lower degree of CIN and tumor heterogeneity than OSCCs, such that they appear as frozen in an early stage of DNA near-diploid aneuploidy, as previously observed for oral preneoplastic lesions. These DI characteristics, which can easily be obtained by hr DNA-FCM, appear to reflect the well-known differences in aggressiveness and prognosis of OVCs and OSCCs. Cancer 2011;117:5052-7. Chronic mechanical trauma/irritation and oral carcinoma: A systematic review showing low evidence to support an association

Case Report: A Peculiar Case of Inflammatory Colitis After SARS-CoV-2 Infection

Summary: We present a case of inflammatory colitis resulting from SARS-CoV-2 infection, leading to the death of a patient with no additional co-morbidity within three weeks of hospitalization. Our findings suggest that SARS-CoV-2 infection can cause immunological alterations, with increased expression of the inhibitory checkpoint PD-1 and its ligand PD-L1 in inflamed mucosal tissue. The presence of the virus in colon tissue was confirmed, and the altered expression of PD-1/PD-L1 indicates a potential link between SARS-CoV-2 infection and an aberrant autoinflammatory response, contributing to early mortality.

Cerivastatin Synergizes with Trametinib and Enhances Its Efficacy in the Therapy of Uveal Melanoma

Combined treatment of trametinib and different drugs targeting YAP/TAZ can overcome resistance. The combined treatment of trametinib and cerivastatin can inhibit the growth of BAP1 mutated and chromosome 3 monosomic uveal melanoma cell lines in vitro and in vivo.

Gingival Overgrowths Revealing PTEN Hamartoma Tumor Syndrome: Report of Novel PTEN Pathogenic Variants

Summary: PTEN hamartoma tumor syndrome (PHTS) is a group of disorders caused by mutations of the PTEN gene, resulting in the development of non-cancerous growths called hamartomas in various parts of the body, including the oral mucosa. PHTS is also associated with an increased risk of malignancies, as PTEN is a tumor suppressor gene that inhibits the progression of several cancers. This study identifies patients with gingival or mucosal enlargements, which led to the diagnosis of PHTS associated with novel PTEN pathogenic variants.

Simulated Microgravity Modulates Focal Adhesion Gene Expression in Human Neural Stem Progenitor Cells

Predictors of malignant transformation in oral leukoplakia and proliferative verrucous leukoplakia: An observational prospective study including the DNA ploidy status

Summary: This prospective observational study examined the factors associated with malignant transformation in localized oral leukoplakia (OL) and proliferative verrucous leukoplakia (PVL). Data on demographics, clinical characteristics, histology, and DNA ploidy status were collected. The presence of OED, DNA ploidy, clinical presentation, and lesion site were found to be associated with malignant transformation in patients with OL, while none of these predictors were associated with PVL. The study suggests that DNA ploidy may be able to identify a subset of OL with low or minimal risk of malignant transformation, but it is not a reliable predictor in PVL patients.
