A gene encoding chitinase was cloned from Ophiocordyceps unilateralis, a Formamidae-specific fungus, collected from Sirindhorn Peat Swamp Forest, Thailand. The O. unilateralis chitinase (OuChi) full-length gene (1311 bp) encodes 436 amino acids with the first 20 amino acids as a putative signal peptide. The gene showed highest identity (78%) to Isaria farinose endochitinase. To investigate if cross-species chitinase expression also enhances fungal toxicity, the mature OuChi gene was subcloned into an Agrobacterium binary vector pPZP-bar and then transformed into Beauveria bassiana strain BCC2659. Chitinase activity was detected using 4-methylumbelliferyl-beta-D-N,N'-diacetylchitobioside. The fungal transformant expressing O. unilateralis chitinase showed higher toxicity against Spodoptera exigua. These results support the hypothesis that chitinolytic enzymes are one of several 'virulence' factors produced by entomopathogenic fungi during host encounter.

Summary: A new class 2 CRISPR-Cas system, Cpf1, optimized from Francisella tularensis subsp. novicida U112, was developed as a versatile platform for precise genome modification in filamentous fungi like Aspergillus aculeatus. The study demonstrated that FnCpf1 efficiently induced site-specific double-strand breaks and insertions or deletions through the endogenous NHEJ DNA repair pathway, highlighting the potential of CRISPR/Cpf1 technology for strain improvements and functional genomics in fungi.

Transcriptomic analysis of the black tiger shrimp (Penaeus monodon) reveals insights into immune development in their early life stages

Pacharaporn Angthong, Tanaporn Uengwetwanit, Sopacha Arayamethakorn, Wanilada Rungrassamee

Summary: The study identified the establishment of the immune system in shrimp during early larval stages through gene analysis. The Toll pathway was found as a baseline immune system established in the nauplius stage, with immune defense mechanisms like pathogen pattern-recognition proteins, antimicrobial peptides, and heat shock proteins established in later larval stages.

Insights Into Transcriptome Profiles Associated With Wooden Breast Myopathy in Broilers Slaughtered at the Age of 6 or 7 Weeks

Yuwares Malila, Tanaporn Uengwetwanit, Krittaporn V. Thanatsang, Sopacha Arayamethakorn, Yanee Srimarut, Massimiliano Petracci, Francesca Soglia, Wanilada Rungrassamee, Wonnop Visessanguan

Summary: The study compared the transcriptomes associated with wooden breast in broilers at two different market ages, revealing differential expression of numerous transcripts in chickens of different ages, particularly impacting metabolism and signaling pathways. Several genes with differential expression related to WB pathology were identified in the FA signaling pathway, indicating the potential involvement of disrupted cell-cell communication in the development of WB myopathy.

Summary: Forage preservation for livestock feeding is commonly achieved through drying the plant material or ensiling it into silage. This study compares fermentation quality and bacterial dynamics in whole-plant corn and Napier grass with and without starter inoculation, and finds that the abundance and diversity of epiphytic LAB in plant raw materials may determine whether LAB starter cultures are necessary for silage fermentation.
