Summary: This study investigated the association of HIF-1 alpha expression and related mechanisms with latent or active tuberculosis infection. The results showed that IL-15 expression was higher in individuals with LTBI, while IL-18 and TNF-alpha expression were similar between LTBI and TB groups. Additionally, patients with active TB had higher serum 25(OH)D3 levels and expression of IL-6, HIF1A, and A20. The study also found that PBMCs from individuals with LTBI showed decreased NF-kappa B phosphorylation and increased TNF-alpha production after HIF-1 alpha blockade.

Summary: E-selectin is a protein expressed on endothelial cells in response to inflammatory cytokines, and it plays a role in leukocyte rolling and extravasation. This study used CRISPR-Cas9 genome editing and NanoLuc Binary Technology (NanoBiT) to tag endogenous E-selectin in human umbilical vein endothelial cells (HUVECs) and monitor its expression in real time. The combination of NanoBiT and CRISPR-Cas9 gene editing provides a powerful tool for monitoring dynamic changes in E-selectin expression on cell surfaces, which can contribute to the discovery of drugs targeting this important inflammatory protein.

Summary: This study investigated the effects of a derivative of glucosamine called DCF001 on the proliferation and chondrogenic induction of stem cells under inflammatory stimuli. The results showed that DCF001 could regulate the expression of CD39, CD73, and TNF receptors, modulate the release of growth factors, and enhance the chondrogenic potential of stem cells. These findings suggest that DCF001 could be a valuable supplement for improving the outcome of cartilage repair using stem cells under inflammatory conditions.
