Summary: Extraction electrospray ionization mass spectrometry (EESI-MS) enables effective ionization of trace molecules in complex samples without sample pretreatment, providing high sensitivity, selectivity, and response speed. It also allows for advanced functionalization of high-energy molecular species. This article systematically reviews the effects of operating conditions on the analytical performance of EESI-MS for various pharmaceutical compounds and introduces its potential application in molecular functionalization for drug discovery.

Summary: In this study, an internal extractive electrospray ionization mass spectrometry (iEESI-MS) method was developed to analyze active metabolites in ginseng samples without pretreatment. A total of 44 metabolites, including 32 ginsenosides, 6 sugars, and 6 organic acids, were identified in the ginseng samples. The study demonstrated that iEESI-MS-based metabolite fingerprints can be used to successfully discriminate different cultivation conditions of ginseng, providing an alternative solution for quality identification of plant drugs.

Summary: The TEMPO-electrocatalyzed acceptorless dehydrogenation of tetrahydroquinoline, which is a green synthesis and hydrogen storage transformation, was investigated by coupling an electrocatalytic system with in situ extraction electrospray ionization mass spectrometry. Dynamic changes in important species and intermediates were monitored, leading to an updated understanding of AD.

Summary: Microdroplet reactions have shown to be an effective method for accelerating large protein reactions, particularly for the digestion, reduction, and deglycosylation of intact antibodies. By introducing appropriate enzymes and reagents into the spray solution, these multiple-step reactions can be completed rapidly, allowing for fast structural characterization of antibodies. This study demonstrates the potential of microdroplets as powerful microreactors for expediting large-molecule reactions and facilitating their structural analysis.

Summary: This study aimed to develop a complete molecular mechanism exploration strategy in the complex medicinal herb decocting system, clarifying the internal molecular mechanism of processing detoxification on Aconitum, and exploring valid approaches for detoxification. By using an advanced online real-time platform, the study successfully characterized components in the complex Aconitum decocting system, discovered that fat meat could detoxify Aconitum directionally while preserving therapeutic effects, and proposed a novel reaction pathway based on nucleophilic substitution mechanism.

Summary: This study demonstrates the use of MALDI-MS imaging for direct measurement of core histone proteoforms from tissues, showing their distribution at near-cellular spatial resolution on sections of human kidney. It provides a new tool for high-throughput accurate identification of proteoforms and mapping biomolecule distributions throughout tissue sections in extended mass ranges.

Summary: The study focuses on improving the mobile phase of electrospray oligonucleotides to reduce spectral complexity and increase sensitivity. By using different combinations of fluorinated alcohols and alkylamines, it is possible to selectively reduce the charge states of oligonucleotides. Additionally, anionic salts are used to stabilize complexes and provide unique structural information about the molecules.

Summary: The study investigated chiral recognition between amino acids and monosaccharides in the gas phase as a model for chemical evolution in interstellar molecular clouds. Results showed that the optical properties of Trp in the clusters induced by 285-nm photoexcitation were useful for identifying and quantifying pentose enantiomers. Additionally, pentose enantiomeric excess in solution could be determined from relative abundances observed in a single product ion spectrum of 285-nm photoexcited hydrogen-bonded clusters of H+(L-Trp) and pentose.

Summary: We developed an immunoassay-based desorption electrospray ionization mass spectrometry imaging (immuno-DESI-MSI) technique for visualizing functional macromolecules. Boronic acid mass tags (BMTs) were synthesized to label antibodies as MSI probes, which can be released from the antibody by cleaving the boronic ester bond under acidic conditions. The BMTs can work in both optical and MS imaging modes and can be readily identified due to the introduction of the boron element.

Summary: A method based on internal extractive electrospray ionization mass spectrometry (iEESI-MS) was developed to characterize chemical components of Panax notoginseng samples grown under different conditions, allowing for rapid molecular characterization and classification of Panax notoginseng. The study identified key characteristic indicators, such as sucrose content, which can be used to discriminate origin, commercial specifications, and cultivation conditions of Panax notoginseng samples. This method shows potential for quality assessment of traditional herbal medicines and pharmaceutical and clinical analysis.

Summary: In this study, a capillary-in-capillary electrospray ionization (CC-ESI) source was developed for spontaneous liquid sampling and quantitative analysis based on the capillary phenomenon and electrospray ionization mass spectrometry (ESI-MS). The CC-ESI had advantages of cheap price, high portability, disposability, quantitation capability, and adequate sensitivity.

Summary: This article introduces a 3D-printable nano-electrospray ionization (nano-ESI) source for Bruker instruments, which can be assembled by anyone with access to a 3D printer. The source can be fitted to any Bruker mass spectrometer with an ionBooster ESI source and only requires minor, reversible changes to the original Bruker hardware. The general utility of the source was demonstrated by recording high-resolution MS spectra of small molecules, intact proteins, as well as complex biological samples in negative and positive ion mode on two different Bruker instruments.

Summary: Electrospray ionization mass spectrometry (ESI-MS) experiments were used to study the solution-like structures of intact proteins, specifically by controlling the protein unfolding process and analyzing folding intermediates. By mixing various pH solutions online with aqueous cytochrome C, the changes in charge states, arrival time distributions (ATDs), and electron capture dissociation (ECD) were measured to detect folding intermediates and unfolded protein structures.

Summary: Exhaled breath analysis provides valuable molecular-level information and has promising potential for precision medicine. However, the lack of validation in multicenter trials hinders the transition of breath tests into routine clinical practice. In this study, an interoperability framework was developed and applied, revealing a technical variability of approximately 20% and identifying a core signature of the human exhaled metabolome. The results showed high inter-subject variability for certain metabolic classes but relative stability in other regions, such as the TCA cycle. This framework and the overview of metabolic coverage will pave the way for future large-scale multicenter trials.

Summary: This study presents quantitative measurements of molecular ions corresponding to diverse aerosol-phase species in indoor air at a time resolution of five seconds using EESI-MS for the first time. The complex spectra of EESI-MS were screened to extract chemical information, demonstrating the method's ability to quantify a large fraction of OA species. Comparisons with other instruments such as AMS and FIGAERO-CIMS showed good correlation and similar signal levels, indicating the effectiveness of EESI-MS in identifying co-emitted gas- and aerosol-phase species.
