Summary: The information provided by SARS-CoV-2 spike (S)-targeting immunoassays is important in clinical-decision making. The performance of two immunoassays, Elecsys (R) Anti-SARS-CoV-2 S assay and LIAISON (R) SARS-CoV-2 TrimericS IgG assay, was compared using sera from vaccinated and unvaccinated individuals. The results showed that the Roche assay had a higher number of positive results and overall sensitivity, and the concordance between the two assays varied depending on the vaccination and infection status.

Summary: G-quadruplexes (G4) are unique nucleic acid structures with diverse conformations. In this study, we developed a novel method, G4-SELEX-Seq, to select an L-RNA aptamer with high binding selectivity to a specific G4 conformation. Our results demonstrate the potential applications of this aptamer in controlling DNA replication and gene expression. This research provides a valuable tool for targeting G4 conformations and regulating G4-mediated biological processes.

Summary: This study investigates the impact of different surface chemistries on the adsorption of proteins to gold nanoparticles. It is found that high-affinity functional groups are essential in preventing protein unfolding and loss of function on the AuNP surface. Additionally, the presence of high-affinity thiol groups enables pH-dependent control of protein orientation and formation of highly active conjugates.

Summary: The present study aimed to determine whether current commercial immunoassays are adequate for detecting anti-Omicron antibodies. The performance of some commercial immunoassays to detect antibodies elicited by Omicron infections was found to be decreased, highlighting the need for optimization to adapt to circulating SARS-CoV-2 strains.

Summary: KRAS gene mutation is common in tumors and has a significant impact on various malignancies. Targeting these mutations is highly desired for cancer therapy. Different strategies, such as covalent binding, protein degradation, protein interaction, salt bridge, and multivalent strategies, have been employed to develop direct inhibitors of KRAS. Several KRAS-directed inhibitors, including FDA-approved drugs sotorasib and adagrasib, as well as specific inhibitors targeting KRAS-G12D and KRAS-G12V, have been developed. These strategies greatly advance the field of KRAS inhibitor development and can also provide insights for targeting other undruggable targets.

Summary: Designing biomolecules with dual photoactivity and photoreactivity remains a challenge. Researchers achieved this by incorporating photocaged tyrosine (pcY) for photoactivity and p-benzoyl-?-phenylalanine (Bpa) for photoreactivity into an epidermal growth factor receptor (EGFR)-targeting antibody fragment, 7D12. They identified a position for incorporating Bpa that had minimal effect on the binding affinity of 7D12 to EGFR. Upon exposure to 365-nm light, this mutant of 7D12 can specifically bind to and form a covalent bond with EGFR.
