Summary: Objective: This experimental study evaluated the therapeutic potential of the Rho-kinase inhibitor hydroxyfasudil in managing allergen-induced asthma. The results showed that treatment with hydroxyfasudil for one week significantly reduced airway resistance and, after 14 days, decreased the number of inflammatory cells in bronchoalveolar lavage fluid and concentrations of cytokines and growth factors in lung tissue.

Summary: Mollugin, derived from Rubia cordifolia L, has been found to possess anti-inflammatory properties. This study aimed to investigate the protective effects of mollugin against shrimp tropomyosin (ST)-induced allergic airway inflammation in mice. The results showed that mollugin reduced eosinophil infiltration, mucus secretion, and lung eosinophil peroxidase activity, as well as the production of Th2 cytokines IL-4 and IL-5. It also inhibited the expression of various genes related to inflammation in the lung tissues. Furthermore, the molecular docking study suggested that mollugin may exert its effects through the inhibition of p38 MAPK and PARP1. Immunohistochemistry analysis revealed that mollugin mitigated arginase-1 expression and macrophage levels in the lungs, supporting its role in inhibiting macrophage polarization.

Summary: This study investigates the potential of OGG1 as a clinically relevant pharmacological target for the treatment of allergic inflammation. The results show that TH5487 treatment decreases goblet cell hyperplasia and mucus production, reduces NF-κB activation, and decreases the expression of proinflammatory cytokines and chemokines, resulting in lower recruitment of immune cells to the lungs. The findings suggest that OGG1 inhibition may have therapeutic effects on allergic asthma.
