Summary: In this study, it was demonstrated that cytosolic HMGB1 induces ferroptosis by binding to ACSL4, exacerbating cellular injury in acute kidney injury (AKI). Inhibition of HMGB1 nucleocytoplasmic translocation and deletion of HMGB1 in tubular epithelial cells reduced AKI, tubular ferroptosis, and inflammation in mice. Co-immunoprecipitation and serial section staining confirmed the interaction between HMGB1 and ACSL4. These findings suggest that cytoplasmic HMGB1 is a regulator of ferroptosis and a promising therapeutic target for AKI.

This study developed a novel nanoparticle drug that can protect the kidneys from IR injury by eliminating hydrogen peroxide (H2O2), exerting anti-inflammatory and anti-apoptotic effects. These nanoparticles have potential therapeutic value not only for renal IR injury but also for other inflammatory diseases associated with reactive oxygen species (ROS).

Moreover, GA improved autophagic clearance and protected against cell injury. These findings suggest that GA may protect against TAC-induced renal injury by improving lysosomal function and regulating autophagy.

Summary: Viral pneumonia is a global health burden, particularly for the elderly and patients with underlying diseases. Recent studies have shown that myeloid-derived suppressor cells (MDSCs) are abundant in these patients, but their role in the progression of viral pneumonia is still unclear. This study found a significant increase in MDSCs in a mouse model and in older mice. When polyinosinic-polycytidylic acid (poly(I:C)) was used to mimic viral pneumonia, mice with renal ischemia/reperfusion (I/R) injury had more severe lung inflammation compared to control mice. Additionally, depleting MDSCs attenuated lung inflammation in mice with I/R injury. Similar results were observed in older mice compared to young mice. Furthermore, transferring in vitro-differentiated MDSCs worsened poly(I:C)-induced lung inflammation. These findings suggest that the increased proportion of MDSCs in mice with renal I/R injury and in older mice exacerbates poly(I:C)-induced lung inflammation.
