Hyper-acetylation contributes to the sensitivity of chemo-resistant prostate cancer cells to histone deacetylase inhibitor Trichostatin A

Therapeutic agents are urgently needed for treating metastatic castration-refractory prostate cancer (mCRPC) that is unresponsive to androgen deprivation and chemotherapy. Our screening assays demonstrated that chemotherapy-resistant prostate cancer (PCa) cells are more sensitive to HDAC inhibitors than paired sensitive PCa cells, as demonstrated by cell proliferation and apoptosis in vitro and in vivo. Kinetic study revealed that TSA-induced apoptosis was significantly dependent on enhanced transcription and protein synthesis in an early stage, which subsequently caused ER stress and apoptosis. ChIP analysis indicated that TSA increased H4K16 acetylation, promoting ER stress gene transcription. The changes in Ac-H4K16, ATF3 and ATF4 were also validated in TSA-treated animals. Further study revealed the higher enzyme activity of HDACs and an increase in acetylated proteins in resistant cells. The higher nucleocytoplasmic acetyl-CoA in resistant cells was responsible for elevated acetylation status of protein and a more vigorous growth state. These results strongly support the pre-clinical application of HDAC inhibitors for treating chemotherapy-resistant mCRPC.

Imaging-Based Body Fat Distribution in Polycystic Ovary Syndrome: A Systematic Review and Meta-Analysis

Summary: A systematic review and meta-analysis showed that women with PCOS have higher accumulations of visceral fat, abdominal subcutaneous fat, total body fat, trunk fat, and android fat compared to BMI-matched controls. However, when only studies using MRI or CT were considered, there were no significant differences in body fat distribution between PCOS and controls. Additionally, young and non-obese PCOS patients were found to be more likely to accumulate android fat.

Summary: This study analyzed the characteristics of the intestinal microbiota in patients with PCOS-IR and explored the mechanism of modified Banxia Xiexin Decoction in treating PCOS-IR. The results showed that PCOS-IR model had an imbalance of intestinal microbiota and Genus Clostridium_sensu_stricto_1 might be the pivotal pathogenic bacteria. Modified Banxia Xiexin Decoction can improve PCOS-IR by regulating the balance of intestinal microbiota.

Summary: Pregnancy-induced hypertension (PIH) is a common pregnancy complication that affects the mother and fetus. The incidence of PIH is higher in pregnancies conceived after assisted reproductive technology (ART) than in spontaneous pregnancies. This study found that using a cytokine profile can effectively predict the risk of PIH in ART pregnancies, providing a basis for early intervention and prevention of PIH.

Distinct cytokine profiles in patients with preeclampsia

Summary: In this study, comprehensive bioinformatic analysis and logistic regression analysis were used to compare serum cytokine profiles between normotensive pregnant subjects and PE patients. The results showed significant differences in cytokine expressions between the two groups, which had significant correlations with the clinical features of PE. A panel of nine cytokines was identified for the diagnosis of PE, and a diagnostic model constructed from these cytokines showed better performance than any individual cytokine, providing a basis for developing early clinical diagnostic and therapeutic strategies for PE.

Summary: This study evaluated the anti-psoriasis effect of Jingfang granules (JFG) and explored its mechanisms using network pharmacology, UPLC-Q-TOF-MS technology, and molecular biotechnology methods. The results showed that JFG improved psoriasis by inhibiting the maturation and activation of dendritic cells and the proliferation and inflammation of keratinocytes. This has important implications for the clinical application of JFG in antipsoriasis therapy.

Refractory Palmoplantar Pustulosis Successfully Treated with JAK Inhibitor Tofacitinib: A Case Series

Summary: This study aims to investigate the efficacy of tofacitinib in refractory palmoplantar pustulosis (PPP). After 4 weeks of treatment, half of the patients achieved more than 80% reduction in PPPASI, and at week 12, 83.3% of the patients had 80% reduction in PPPASI with no serious adverse events reported. Therefore, tofacitinib shows promise as a treatment for refractory PPP, but further clinical observation and research are needed.

Polycystic ovary syndrome and 25-hydroxyvitamin D: A bidirectional two-sample Mendelian randomization study

Summary: This study used a bidirectional two-sample Mendelian randomization (MR) design to investigate the causal association between polycystic ovary syndrome (PCOS) and 25-hydroxyvitamin D (25OHD) deficiency. The findings showed a negative association between genetically determined PCOS and 25OHD levels, but this causal effect disappeared when adjusting for obesity and insulin resistance. The study failed to find a substantial causal effect of 25OHD deficiency on the risk of PCOS. Further observational studies and clinical trials are needed.
