Summary: Targeting the Plasmodium proteasome is a promising strategy to combat malaria, especially with the increasing reports of resistance to current therapies. In this study, we conducted structure-activity relationship studies on a macrocyclic scaffold and identified compound TDI-8414, which showed potent antiparasitic activity, high selectivity, improved solubility, and enhanced metabolic stability.

Highly selective DNA aptamers were successfully identified and adapted into structure-switching aptamer fluorescent sensors for drug detection in human serum and crushed tablets. The sensor platform offers a rapid, inexpensive method for tracking antimalarial drug use and quality, with potential for detecting other small-molecule drugs.

Summary: Most malaria deaths in sub-Saharan Africa occur in children, so it is crucial for this age group to have access to appropriate treatment and dosage. However, the recommended dosage of artemether-lumefantrine has been found to cause underexposure or overexposure in some children. This article aimed to estimate doses that mimic adult exposure, using physiological information from children and limited pharmacokinetic data from adults. The results showed that some children were underexposed and others were overexposed, which can lead to treatment failure and toxicity. Clinically efficacy of the suggested doses should be further studied.

Summary: Recently, the antitumor activity of artemisinin and its derivatives has been discovered. This study constructed novel Pt-IV-artesunate complexes to combine the antitumor advantages of artesunate and platinum drugs. The derivatives, especially 10f, exhibited potent in vitro antitumor activities against various cancer cell lines. Compound 10f demonstrated remarkable in vivo antitumor efficacy in a xenograft model, and also showed potent in vivo antimalarial activity. This study highlights the therapeutic potential of Pt(IV)-artesunate complexes as antitumor and antimalarial agents.

Summary: The rise in Plasmodium falciparum resistance to dihydroartemisinin-piperaquine in Vietnam has led to the evaluation of alternative artemisinin-based combination therapies. A trial of pyronaridine-artesunate in Dak Nong province, Vietnam showed high efficacy in treating falciparum malaria, despite a high prevalence of parasites with resistance mutations to both artemisinins and piperaquine among the patients.
