Summary: Chronic lymphocytic leukemia (CLL) cells efficiently evade immunosurveillance through close interaction with stromal niches in the bone marrow and lymph nodes. Enhanced PD-L1 expression on CLL cells following stromal contact contributes to immune escape, controlled by a Notch-c-Myc-EZH2 signaling axis. Elevated PD-L1 levels confer resistance towards activated autologous T-cells, highlighting potential therapeutic targets for improving immune responses in CLL patients.

Anticancer Aminoferrocene Derivatives Inducing Production of Mitochondrial Reactive Oxygen Species

Summary: Elevated levels of reactive oxygen species (ROS) and deficient mitochondria are weak points of cancer cells. Designing highly potent anticancer drugs that target both ROS and mitochondria is a valid therapeutic strategy. However, limiting the drug effects to cancer cells without affecting normal ones remains a challenge. In this study, we developed novel aminoferrocene derivatives that are chemically stable in the presence of ROS, generate mitochondrial ROS in cancer cells but not normal cells, and exhibit anticancer effects in vivo.

The metabolic profile of reconstituting T-cells, NK-cells, and monocytes following autologous stem cell transplantation and its impact on outcome

Summary: This study found that reconstituting immune cells undergo metabolic changes after autologous stem cell transplantation, including increased proliferative activity and upregulation of glycolytic and fatty acid oxidation machinery. This metabolic activation is more pronounced in T-cells of advanced differentiation stages, CD56(bright) NK cells, and CD14(++)CD16(+) intermediate monocytes. Furthermore, persistent metabolic activation is associated with poor disease course in lymphoma patients within the first year following transplantation.

Summary: This study reveals that CLL cell-derived EVs impair the viability, proliferation, activation, and metabolism of T-cells, while promoting T-cell exhaustion and the formation of regulatory T-cell subsets. The presence of immunological checkpoints (ICs) loaded on CLL-EVs may explain the T-cell dysregulations observed in CLL patients.

Summary: T-cell-based approaches are increasingly important in cancer treatment, and the relationship between immune cell metabolism and immunotherapy success is crucial. Excessive exposure of T cells to fatty acids, known as lipotoxicity, can have detrimental effects in the context of an altered lipid tumor microenvironment. Understanding T cell-related lipotoxicity and developing potential therapeutic approaches is of great importance.

Summary: The insertion site of ITDs in the FLT3 gene influences the sensitivity to TKIs therapy in AML. The study identifies the WEE1-CDK1 axis as a conserved role in TKIs resistance and shows that inhibition of WEE1 kinase enhances the pro-apoptotic effect of TKIs therapy. Therefore, combination therapy with WEE1 inhibitor and TKI may improve the clinical outcome of AML patients.

Summary: Allo-SCT is a curative treatment for hematologic malignancies, relying on the graft-versus-leukemia (GVL) effect of donor T cells. Oxidative stress caused by DNA damage weakens the function of T cells and hampers the GVL effect. High levels of 8-hydroxydeoxyguanosine (8-OHdG), a biomarker for oxidative stress, were associated with enhanced T-cell activation, premature exhaustion, increased relapse risk, and shorter overall survival. Enhancing DNA repair in reconstituting T cells could improve GVL efficacy.

Summary: Allogeneic hematopoietic stem cell transplantation (allo-HSCT) is the only curative treatment option for certain hematological malignancies. However, allo-HSCT can also lead to graft-versus-host disease (GvHD) and disease relapse. Recent studies have found that extracellular vesicles (EVs) expressing PD-L1 can suppress T-cell responses and contribute to GvHD and disease relapse in allo-HSCT patients.
