Designing lipid nanoparticles for topical ocular drug delivery

Topically-applied dosage forms, such eye drops, are the most used formulations in the treatment of ocular diseases. Nonetheless, because of the special protection of the eye associated with the ocular surface, drug bioavailability and subsequent therapeutic efficiency obtained with these conventional dosage forms are very low. Recently, novel drug delivery systems have been proposed to solve the main drawbacks of conventional formulations. Nanotechnology, and more specifically lipid nanoparticles, have emerged as promising modified eyedrops with the purpose of improving therapeutic efficiency without compromising drug safety and patient compliance. The purpose of this review is to offer an overview of lipid nanoparticles as feasible alternatives to the conventional topically-applied dosage forms. We discuss the main limitations of topical ocular drug delivery and describe how correctly designed lipid nanoparticles can be highly valuable tools to overcome the constraints imposed by the ocular surface. Special emphasis is placed on the description of production methods and bulk materials used in the development of lipid nanoparticles for ophthalmic use and how both issues will determine the physicochemical and biopharmaceutical properties of the developed nanosystems.

Epithelial cells removed in advanced surface ablation (ASA) surgery can be used as a source of corneal samples to perform in vitro studies

Summary: Samples obtained from corneal surface ablation procedures can be used for in vitro studies, as cells showed high viability and remained viable for up to 14 days in culture. Protein expression analysis confirmed the presence of cytokeratin-3, ZO-1, Ki67, and E-cadherin, while gene expression studies showed successful isolation of RNA and detection of GAPDH expression.

Summary: A green technique was developed to extract hyaluronic acid (HA) from tuna vitreous humor (TVH) for potential use in managing dry eye disease. The extracted HA displayed antioxidant effects and showed potential as a compound in dry eye disease management. The technique also produced deep eutectic solvents (DES) that exhibited efficient antimicrobial properties.

Improved ocular delivery of quercetin and resveratrol: A comparative study between binary and ternary cyclodextrin complexes

Summary: This study compared the effects of complexes with cyclodextrins and tertiary complexes with the addition of hyaluronic acid on the solubility and stability of quercetin and resveratrol. The results showed that both complexes improved the solubility and stability of the drugs, and had no cytotoxic effects. These findings indicate the potential use of these complexes as topical treatment for dry eye disease.

Characterization and functional performance of a commercial human conjunctival epithelial cell line

Summary: In this study, a new protocol was developed to culture IM-HConEpiC cells without collagen-coated surfaces, and the cells were found to maintain a conjunctival epithelial phenotype. The study also demonstrated that the cells responded appropriately to oxidative stress and inflammatory stimuli, indicating that they can be a valuable tool for functional studies of the conjunctiva.

Summary: This study aimed to investigate the loading of sildenafil citrate in three commercial transdermal vehicles using 3D full-thickness skin equivalent and compare the results with permeability experiments using porcine skin. The results showed that the results obtained using the 3D skin equivalent were comparable to those obtained using porcine skin, suggesting that the 3D skin model can be a valid alternative for ex-vivo skin absorption experiments.
