This article provides a comprehensive understanding of osteoporosis, including its causes, effects on bone density, and the role played by genetic pathways and gut flora. It investigates the link between gut flora and osteoporosis through animal and human experiments. Additionally, it speculates on possible developments in osteoporosis prevention and therapy.
Osteoporosis is a systemic condition of the skeleton that leads to diminished bone mass, a breakdown in the bone tissue's microscopic architecture, and an elevated risk of breaking a bone. The elderly and women particularly after menopause are disproportionately affected, and the condition generally stays undiagnosed until a broken bone causes severe pain and immobility. Causes of osteoporosis include low bone mass, more than normal bone loss, changes in hormone levels (decreased estrogen or testosterone), certain diseases and therapies, and lifestyle factors like smoking and inactivity. The spine, hip, and forearm are particularly vulnerable to osteoporosis-related fractures. The purpose of this article is to present a thorough understanding of osteoporosis, including the disease's connection to bone density in humans, and the major part played by genetic pathways and gut flora. The causes of osteoporosis, the effects of aging on bone density, and why some groups experience a higher incidence of the disease than others are investigated. The paper also includes animal and human experiments investigating the link between gut flora and osteoporosis. Finally, it looks to the future and speculates on possible developments in osteoporosis prevention and therapy.

Comparative accounts of probiotic properties of spore and vegetative cells of Bacillus clausii UBBC07 and in silico analysis of probiotic function

J. J. Ahire, M. S. Kashikar, R. S. Madempudi

Summary: The study demonstrated that B. clausii UBBC07 spores exhibit higher survival and adhesion in vitro gastrointestinal conditions compared to vegetative cells, attributed to their stability and hydrophobicity. In addition, the identification of genome features suggested their contribution to probiotic properties.

In Vitro Assessment of Lactobacillus crispatus UBLCp01, Lactobacillus gasseri UBLG36, and Lactobacillus johnsonii UBLJ01 as a Potential Vaginal Probiotic Candidate

J. J. Ahire, S. Sahoo, M. S. Kashikar, A Heerekar, S. G. Lakshmi, R. S. Madempudi

Summary: In this study, lactobacillus strains isolated from healthy reproductive age Indian women were screened for beneficial probiotic properties. The strains showed survival capabilities under various conditions, inhibited the growth of pathogens, and could adhere and co-aggregate with them. They also secreted exopolysaccharides and formed biofilms. Safety assessment indicated their potential for human trials to prevent/treat vaginal dysbiosis.

Effect of Bacillus coagulans Unique IS2 with Lactulose on Functional Constipation in Adults: a Double-Blind Placebo Controlled Study

R. Venkataraman, R. Shenoy, J. J. Ahire, J. Neelamraju, R. S. Madempudi

Summary: This study evaluated the efficacy of combining Bacillus coagulans Unique IS2 and lactulose in treating functional constipation in adults. The results showed that the combination of B.coagulans Unique IS2 and lactulose was more effective than lactulose alone in relieving constipation symptoms in a shorter period of time.

Survival of Limosilactobacillus reuteri UBLRu-87 during passage through the in vitro gut model system

J. J. Ahire, N. U. Mokashe, M. S. Kashikar, R. S. Madempudi

Summary: This study investigated the survival of Limosilactobacillus reuteri UBLRu-87 in an in vitro gut model system. The results showed that without glucose, the strain lost its ability to grow and multiply in the intestines, but remained viable in a non-culturable state. In the presence of glucose, the strain was able to protect itself and enhance its viability during passage through the gastrointestinal tract.

Riboswitch-mediated regulation of riboflavin biosynthesis genes in prokaryotes

Vikram, Vijendra Mishra, Ananya Rana, Jayesh J. Ahire

Summary: Riboswitches are used by prokaryotic organisms to quantify intracellular metabolite concentration. They control gene expression by binding with specific metabolites and modulating various biochemical pathways. This article focuses on how riboswitch regulates the riboflavin biosynthesis pathway in Bacillus subtilis and Lactobacillus plantarum.

Genetic and phenotypic assessments for the safety of probiotic Bacillus clausii 088AE

Dina B. Saroj, Jayesh J. Ahire, Rohit Shukla

Summary: In this study, the authors analyzed the whole genome sequence of the probiotic Bacillus clausii 088AE and identified its genome features that contribute to its probiotic properties. The genome sequence had a single scaffold of 4,598,457 bp with 44.74 mol% G + C and was annotated to have 4371 coding genes, 75 tRNAs, and 22 rRNAs. Taxonomic analysis revealed its high similarity (99%) to B. clausii DSM 8716. The presence of genome features related to safety, genome stability, and probiotic characteristics confirmed the strain to be safe for use as a probiotic.

Summary: In this study, the production, characterization, and identification of antimicrobial compounds from Alkalihalobacillus clausii 088AE were evaluated. The results demonstrated that the supernatant of 48-h-old A. clausii can inhibit the growth of Micrococcus luteus (7.0 & PLUSMN; 0.51 mm). Enhanced antimicrobial activity (19.67 & PLUSMN; 0.58 mm) against indicator strain was observed after cultivating 088AE on solid media with XAD16N beads for 5 days and extracting the antimicrobial compound with isopropanol. The purified compound, clausin, showed stability in various conditions and antimicrobial activity against Gram-positive pathogens.
