Summary: Baihu-Guizhi decoction (BHGZD), a traditional Chinese medicine-originated prescription, has shown promising therapeutic effects in the treatment of rheumatoid arthritis (RA) by modulating the TLR4-mediated inflammasome activation. BHGZD not only alleviated RA symptoms in animal models but also inhibited pyroptosis and inflammatory signaling pathways in vitro, suggesting its potential as an immunomodulatory and anti-inflammatory agent in RA treatment.

Summary: The NLRP3 inflammasome is a cytosolic multiprotein complex that responds to microbial infection, endogenous danger signals, and environmental stimuli. Its activation can induce pyroptosis and release of inflammatory cytokines, contributing to innate immune defense. However, aberrant activation of the NLRP3 inflammasome is associated with inflammatory diseases, and there is a close relationship between cell death pathways and NLRP3 inflammasome activation.

Summary: Pseudorabies virus (PRV) preferentially invades neural tissue and organs, causing multisystemic lesions. In this study, it was shown that PRV infection triggers GSDMD-mediated pyroptosis in porcine alveolar macrophage cells through the activation of caspase-1 and the cleaving of GSDMD. NLRP3 and IFI16 inflammasomes were also activated, contributing to pyroptosis during PRV infection. These findings enhance our understanding of the inflammatory response and cell death pathways caused by PRV.

Summary: Increasing evidence suggests that neuroinflammation may be a contributing factor to the failure of clinical antidepressants. The NLRP3 inflammasome, a multiprotein complex, has been identified as a key player in the development of neuroinflammation. Inhibition of NLRP3 inflammasome activation presents a promising therapeutic strategy for depression. This review summarizes current research on the role of NLRP3 inflammasome in depression pathology, including its activation in patients and animal models, potential driving mechanisms, and its pathogenetic role in depression. Additionally, it provides an overview of current and potential antidepressants targeting NLRP3 inflammasome.

Summary: Programmed cell death (PCD) is a cell death mechanism that depends on specific genes encoding signals or activities. PCD includes apoptosis, pyroptosis, autophagy, and necrosis (programmed necrosis). Among these mechanisms, pyroptosis is mediated by the gasdermin family and is accompanied by inflammatory and immune responses. The relationship between pyroptosis and cancer is complex and its effects vary depending on the tissue and genetic background.

Summary: Allergic rhinitis (AR) is a chronic inflammatory disease caused by immunoglobulin E-mediated immune response, which has been increasing in incidence and significantly impacting individuals' wellbeing. Due to the complex pathogenesis of AR, treatment outcomes are still not ideal. Recent studies have found that NLRP3 inflammasome-mediated pyroptosis is widely involved in the occurrence and development of AR through various pathways. This article reviews the mechanism of pyroptosis and its research progress in AR, proposing potential therapeutic targets for innovative management approaches.
