Summary: Individuals carrying the PCSK9(Q152H) variation have low cholesterol levels and normal liver function despite retaining the uncleavable form of PCSK9 in liver cells. Overexpression of PCSK9(Q152H) in mice unexpectedly protected against ER stress and liver injury, suggesting a potential hepatoprotective role of this variant. This study provides insights into the protective effects of PCSK9(Q152H) on liver function and suggests implications for ER stress-related conditions.

Summary: PCSK9, a key regulator of LDL cholesterol metabolism, is found to be highly expressed in undifferentiated hiPSCs. Inhibition of PCSK9 in hiPSCs leads to decreased phosphorylation of SMAD2 and cell proliferation, while overexpression of PCSK9 stimulates cell proliferation by regulating the expression of DACT2, an endogenous inhibitor involved in TGF beta-R1 lysosomal degradation. This highlights a new role of PCSK9 in cellular proliferation and development.

Summary: The spike protein of SARS-CoV-2 plays a crucial role in infecting the lungs and other tissues by binding to the ACE2 receptor. Cleavage of the spike protein at S1/S2 and S2' sites is important for viral entry and fusion. This study identifies the fusion activation site S2' as KPSKR815 down arrow and demonstrates that its cleavage is enhanced by ACE2 engagement. The combination of furin and TMPRSS2 inhibitors shows promise as potent antivirals against SARS-CoV-2.

Summary: In this study, the regulatory miRNAs of PCSK7 gene expression were predicted using bioinformatics algorithms. Four miRNAs were identified as potential regulators of PCSK7 expression. The experimental results confirmed the interactions between these miRNAs and PCSK7, and revealed their regulatory effects on PCSK7 expression levels.
