Role of Telomeres Shortening in Atherogenesis: An Overview

The shortening of telomeres is associated with cell senescence and the development of atherosclerosis, while the expression of telomerase can elongate telomeres and resist senescence. Atherosclerosis is believed to be linked to local cell senescence, with wall shear stress in the arteries possibly playing a role.
It is known that the shortening of the telomeres leads to cell senescence, accompanied by acquiring of pro-inflammatory phenotype. The expression of telomerase can elongate telomeres and resist the onset of senescence. The initiation of atherosclerosis is believed to be associated with local senescence of the endothelial cells of the arteries in places with either low or multidirectional oscillatory wall shear stress. The process of regeneration of the artery surface that has begun does not lead to success for several reasons. Atherosclerotic plaques are formed, which, when developed, lead to fatal consequences, which are the leading causes of death in the modern world. The pronounced age dependence of the manifestations of atherosclerosis pushes scientists to try to link the development of atherosclerosis with telomere length. The study of the role of telomere shortening in atherosclerosis is mainly limited to measuring the telomeres of blood cells, and only in rare cases (surgery or post-mortem examination) are the telomeres of local cells available for measurement. The review discusses the basic issues of cellular aging and the interpretation of telomere measurement data in atherosclerosis, as well as the prospects for the prevention and possible treatment of atherosclerosis.

Hypotheses on Atherogenesis Triggering: Does the Infectious Nature of Atherosclerosis Development Have a Substruction?

Summary: Since the end of the 20th century, it has been recognized that atherosclerosis is an inflammatory disease and the trigger for inflammation in the vascular walls is still uncertain. Numerous hypotheses have been proposed to explain the causes of atherogenesis, all supported by strong evidence. These causes include lipoprotein modification, oxidative transformation, shear stress, endothelial dysfunction, free radicals' action, homocysteinemia, diabetes mellitus, and decreased nitric oxide level. The latest hypothesis suggests the infectious nature of atherogenesis, with bacterial and viral infections possibly playing a role. This paper analyzes the existing hypotheses for atherogenesis and focuses on the contribution of bacterial and viral infections in the pathogenesis of atherosclerosis and cardiovascular disease.

Summary: Despite the disadvantages of classical statin therapy, including side effects and difficulty with compliance, modern and more efficient therapeutic strategies are needed for cardiovascular disease treatment. PCSK9 has been identified as a promoter of atherogenesis, making it a target for new treatment methods that can prevent or alleviate atherosclerosis. This review summarizes the role of PCSK9 in the development and progression of atherosclerosis and considers existing PCSK9 inhibitors.

Creation of Mitochondrial Disease Models Using Mitochondrial DNA Editing

Summary: Mitochondrial diseases are human hereditary diseases characterized by mitochondrial dysfunction and disrupted cellular energy synthesis. Animal models are the most suitable and promising models for studying these diseases, although they are rare and difficult to develop due to challenges in genetic editing of mitochondrial DNA. This review discusses the animal models of mitochondrial diseases and the recently developed approaches used to create them, as well as the studies on mitochondrial diseases and metabolic disorders caused by mitochondrial DNA mutations.

Many studies have shown that low levels of HDL cholesterol are associated with increased cardiovascular risk. Therefore, HDL-C is considered a promising therapeutic target. This review discusses current and potential HDL-based therapeutic strategies.

Some heat shock proteins have antiatherosclerotic effects, while others' roles are unclear. This review focuses on the antiatherosclerotic activity of specific heat shock proteins.

Future research on EOs has the potential to discover new bioactive compounds and develop effective agents for the treatment of atherosclerosis and related diseases such as diabetes, metabolic syndrome, and obesity.
