5-Fluorouracil efficacy requires anti-tumor immunity triggered by cancer-cell-intrinsic STING

5-FU reduces tumor burden by triggering anti-tumor immunity through activating cancer-cell-intrinsic STING, which requires the involvement of cGAS and interferons. The presence of T cells is also crucial for the efficacy of 5-FU treatment.
5-Fluorouracil (5-FU) is a widely used chemotherapeutic drug, but the mechanisms underlying 5-FU efficacy in immunocompetent hosts in vivo remain largely elusive. Through modeling 5-FU response of murine colon and melanoma tumors, we report that effective reduction of tumor burden by 5-FU is dependent on anti-tumor immunity triggered by the activation of cancer-cell-intrinsic STING. While the loss of STING does not induce 5-FU resistance in vitro, effective 5-FU responsiveness in vivo requires cancer-cell-intrinsic cGAS, STING, and subsequent type I interferon (IFN) production, as well as IFN-sensing by bone-marrow-derived cells. In the absence of cancer-cell-intrinsic STING, a much higher dose of 5-FU is needed to reduce tumor burden. 5-FU treatment leads to increased intratumoral T cells, and T-cell depletion significantly reduces the efficacy of 5-FU in vivo. In human colorectal specimens, higher STING expression is associated with better survival and responsiveness to chemotherapy. Our results support a model in which 5-FU triggers cancer-cell-initiated anti-tumor immunity to reduce tumor burden, and our findings could be harnessed to improve therapeutic effectiveness and toxicity for colon and other cancers.

Evitar: designing anti-viral RNA therapies against future RNA viruses

Summary: This study developed the Evitar pipeline for designing anti-viral siRNAs and CRISPR Cas13a gRNA sequences. Time simulations using known coronavirus genomes showed that pre-designed siRNAs and gRNAs could target SARS-CoV-2 and other viruses. In vitro experiments confirmed the potential of designed siRNAs to suppress SARS-CoV-2 viral sequences. The results support the utility of Evitar in pre-designing therapeutics against future RNA virus outbreaks.

Epigenetically suppressed tumor cell intrinsic STING promotes tumor immune escape

Summary: DNA sensing through the cGAS-STING pathway is important in cancer immunosurveillance. Activation of STING in the tumor environment is an attractive approach to induce anti-tumor immunity, but it is limited due to epigenetic silencing of STING in many tumors. In this study, the inhibition of KDM5 family histone demethylases restored STING expression in breast cancer cells and activated the cGAS-STING pathway, showing the potential of KDM5 inhibitors as novel immune modulators in cancer therapies.

The Crossroads of Cancer Epigenetics and Immune Checkpoint Therapy

Summary: Immune checkpoint inhibitors (ICI) have shown significant improvements in treating various cancers, but there are still challenges to overcome. Some cold tumor types, like pancreatic cancer, have low response rates to ICI due to low immunogenicity. Additionally, patients who initially respond to ICI may experience T-cell exhaustion, leading to a lack of sustained response. Recent studies have highlighted the role of epigenetic modifiers in regulating tumor cell immunity and T-cell exhaustion, suggesting that targeting the intersection of epigenetics and immune checkpoint therapy could enhance antitumor immune responses.

Global, regional, and national incidence and prevalence of systemic sclerosis

Summary: In this study, we estimated the unbiased epidemiological data of SSc globally and in specific countries through systematic searches of epidemiological studies. The results showed that 83.9% of countries worldwide lack epidemiological data on SSc. The global incidence and prevalence of SSc were estimated to be 8.64 per 100,000 person-years and 18.87 per 100,000 persons, respectively. There is a higher incidence and prevalence of SSc in females, adults, and high-income level countries. It is necessary to conduct large-scale and standardized epidemiological investigations of SSc, especially in low- and middle-income countries.

Summary: Epigenomic-wide DNA methylation profiling has the potential to reflect risks associated with electronic cigarette exposure and individual poor health outcomes. Our study in male ApoE-/- mice reveals significant alterations in CpG sites after electronic cigarette exposure, with a high percentage annotated with known genes. Pathway analysis indicates activation of MAPK and cardiomyocyte-related signaling pathways, suggesting potential cell-damaging effects of prolonged e-cigarette inhalation.
