Summary: As the sister group to bilaterians, cnidarians provide valuable insights into evolutionary aspects of animal development, physiology, and behavior. This study introduces a gene targeting siRNA electroporation method that enables the knockdown of target genes in fertilized eggs of jellyfish species. The method has been successfully applied to knock down endogenous genes in Clytia hemisphaerica and Cladonema paciificum, resulting in phenotypic changes related to gene expression. The development of this technique holds promise for future research on animal development.

Summary: This study demonstrates the visualization of Cas9 protein in embryos after electroporation and its correlation with the genome editing rate of the offspring using a fluorescent Cas9 fusion protein. The data suggest that the TAKE method using fluorescently-labeled nucleases can be used to optimize the delivery conditions and verify nuclease delivery into individual embryos for the efficient production of genome-edited animals.

Summary: Most colonial marine invertebrates have the ability to distinguish between themselves and conspecifics, known as allorecognition. In the cnidarian Hydractinia symbiolongicarpus, allorecognition is controlled by at least two genes, Allorecognition 1 (Alr1) and Allorecognition 2 (Alr2), which encode highly polymorphic cell-surface proteins. These proteins are members of the immunoglobulin superfamily (IgSF), with V-set and I-set Ig domains unlike any previously identified in animals. The discovery of these genes expands our understanding of invertebrate histocompatibility.

Summary: The chemical modification of DNA called 6mA randomly accumulates in early embryos of Hydractinia. Knocking down Alkbh1, a protein involved in the clearance of 6mA, leads to higher levels of 6mA and delayed activation of zygotic transcription. This suggests that 6mA inhibits gene expression in early embryonic genomes, and defects in 6mA clearance may be linked to certain pathologies.
