Summary: This study revealed that Ginsenoside Rh2 (GRh2) has anti-inflammatory and antioxidant effects, and can reduce the release of myocardial enzymes and production of inflammatory cytokines caused by myocardial ischemia/reperfusion (I/R) injury. GRh2 also decreased the area of myocardial infarction, improved histological changes in the myocardium, and enhanced cardiac function. Furthermore, GRh2 reduced oxidative stress and inflammation in cardiomyocytes after reperfusion by regulating the Nrf2/HO-1/NLRP3 signaling pathway.

Summary: Ferroptosis is involved in the development of neurological disorders, and regulating ferroptosis shows potential therapeutic effects in nervous system diseases. The study identified ATP2B3 as a target protein through TMT-based proteomic analysis, and inhibiting ATP2B3 alleviated erastin-induced ferroptosis in HT-22 cells by modulating the P62-KEAP1-NRF2-HO-1 pathway. Other interventions targeting related proteins also reversed the effects of ferroptosis. Overall, this study highlights the importance of ATP2B3 inhibition in mitigating ferroptosis in HT-22 cells.

Summary: The study found that multi-walled carbon nanotubes (MWCNTs) induce oxidative stress by overproducing reactive oxygen species (ROS), leading to cytotoxicity. However, celastrol, a plant-derived compound, has antioxidant effects by reducing ROS production. The results showed that MWCNT exposure decreased cell viability and increased ROS production. Knockdown of Nrf2 increased ROS production and cytotoxicity, while knockdown of Keap1 decreased ROS production and attenuated cytotoxicity. Treatment with celastrol significantly reduced ROS production and promoted Keap1 degradation, leading to increased Nrf2 translocation to the nucleus and increased HO-1 expression. In vivo results demonstrated that celastrol alleviated inflammatory damage in lung tissues, increased levels of antioxidants, GSH and SOD, and promoted HO-1 expression in MWCNT-treated mice. Therefore, celastrol can alleviate MWCNT-induced oxidative stress through the Keap1/Nrf2/HO-1 signaling pathway.

Summary: In this study, the effects of ShenKang Injection (SKI) on the kidneys of DKD rats and its effect on oxidative stress mediated by the Keap1/Nrf2/Ho-1 signaling pathway were investigated through network pharmacology and in vivo and in vitro experiments. It was found that SKI can protect kidney function in DKD rats, delay DKD progression, and inhibit AGEs-induced oxidative stress damage in HK-2 cells. The mechanism of SKI in improving DKD may be achieved by activating the Keap1/Nrf2/Ho-1 signal transduction pathway.

Summary: Epilepsy is a chronic disease that affects a wide range of people, and one-third of patients do not respond to antiepileptic drugs. Oxidative stress has been recognized as playing a role in acquired epilepsy, and targeting oxidative stress with adjuvant antiepileptic drugs may be a new therapeutic strategy. This study reveals the resistance of ginsenoside Rh2 to oxidative stress induced by epileptic activity and identifies the overexpression of FOXO3a in epilepsy tissue, which leads to more severe oxidative stress. In addition, the study uncovers the involvement of the KEAP1-NRF2 pathway in regulating oxidative stress and demonstrates the ability of ginsenoside Rh2 to inhibit the activation of FOXO3a, thus providing potential targets for the development of new treatments for epilepsy.
