The lncRNA CARMEN is known to direct the specification of cardiomyocytes (CMs) and smooth muscle cells (SMCs), and generates multiple alternatively spliced isoforms. This study identifies an isoform, CARMEN-201, as crucial for SMC commitment by interacting with a transcriptional repressor and blocking the CM gene program. These findings highlight the importance of alternative splicing and functional transposable elements in lncRNAs during heart development.
