Summary: Acute Lung Injury (ALI) and Acute Respiratory Distress Syndrome (ARDS) are major causes of ICU death, characterized by severe hypoxemia and inflammation leading to poor lung compliance. Long non-coding RNA (lncRNA) has been shown to be abundantly expressed in ALI and targeting lncRNAs can alleviate ALI. Therefore, lncRNAs hold promise as potential therapeutic targets for ALI.

Summary: This study analyzed the transcriptome data of octoploid strawberry fruits treated with ABA and its inhibitor NGDA to identify responsive LncRNAs and coding genes. It identified several high-confidence LncRNAs specific to the ABA pathway and 22 hub-responsive LncRNAs, providing new insights into fruit ripening in strawberry or other Rosaceae crops.

Summary: This study identified a five immune-related lncRNAs signature that can accurately predict the prognosis of prostate adenocarcinoma patients. Further analysis showed that the signature can stratify patients into high-risk and low-risk groups, and gene expression analysis indicated significant up-regulation of four lncRNAs in tumor tissues. These findings suggest the clinical significance of the signature in prognosis prediction and immunotherapy for prostate adenocarcinoma patients.

Summary: Long non-coding RNAs (lncRNAs) have important regulatory roles in tissue morphology and functions. This study constructed comprehensive transcriptomic atlases of human lncRNAs in normal and disease states, revealing high consistency within the same tissues and complexity in specialized tissues. Tissue-elevated (TE) lncRNAs were identified and shown to have important roles in tissue-related pathways. The expression features of TE lncRNAs made them effective biomarkers for tissue classification and were associated with cancer and patient survival.

Summary: Non-alcoholic fatty liver disease (NAFLD) is the most common chronic liver disease worldwide. Long non-coding RNA (lncRNA) and circular RNA (circRNA) have emerged as potential non-invasive biomarkers for predicting NAFLD. This systematic review highlights the potential of lncRNA and circRNA, such as NEAT1, MEG3, MALAT1, and SCAR, for early diagnosis and progression assessment of NAFLD, NASH, and fibrosis. Large-cohort studies and further exploration of the functional mechanisms are needed to validate their clinical value.

Summary: An effective loss-of-function study is necessary to investigate the biological function of lncRNA, and CRISPR-based genome editing is the most widely used method for inactivating lncRNA function at the genomic level. However, the complex genomic interplay between lncRNA and neighbor genes makes it challenging to accurately interpret lncRNA function. This article discusses the advantages and disadvantages of different lncRNA knockout methods and envisions potential future directions for lncRNA functional study.
