Summary: In this study, a soluble NKCAR was designed to trigger NK cell cytotoxicity by binding with the ligand MICA through the NKG2D receptor expressed on NK cells. The NKCAR showed bifunctional activity, recognizing both the CD20 antigen of target cells and the NKG2D receptor of NKL cells. The soluble NKCAR mediated the killing of CD20(+) Daudi cells in vitro, indicating its potential clinical application for killing target tumor cells.

Summary: Natural killer cells can eliminate tumors through direct cytotoxicity and antibody dependent cellular cytotoxicity. The use of antibodies targeting specific surface receptors can enhance antibody dependent cellular cytotoxicity. CD16 receptors may mediate both direct cytotoxicity and antibody dependent cellular cytotoxicity, leading to competitive usage depending on differentiation status of tumor cells and activation stage of NK cells.

Summary: This study explores the effect of high-fat diet on the development of renal cell carcinoma (RCC) and the changes in immune cell groups. The findings reveal that high-fat diet impacts tumor growth and alters the frequencies of T cells, B cells, and NK cells. These findings provide new insights for immunotherapy in individuals with obese RCC.

Summary: Abnormal intracellular metabolism in pancreatic cancer not only provides nutrition for tumor development, but also affects the function of immune cells in the immune microenvironment, promoting immune escape. This review discusses the emerging role of immune cells in pancreatic cancer, focusing on metabolic reprogramming and key metabolic pathways. The hotspots of immune cell metabolic reprogramming in pancreatic cancer mainly involve glucose metabolism, lipid metabolism, tricarboxylic acid cycle, and amino acid metabolism, which impact the function of anti-tumor immune cells and immunosuppressive cells through key metabolic signaling pathways. The review also elaborates on drugs targeting tumor metabolism pathways for clinical treatment of pancreatic cancer.

Summary: The field of immuno-oncology has revolutionized cancer patient care, but there are still challenges with current therapeutic approaches targeting T cells. Engaging the innate immune system, especially natural killer (NK) cells, has emerged as a promising modality in immunotherapy. This review focuses on therapeutic strategies that selectively engage NK cells for cancer therapy, particularly targeting the activating receptors NKG2D and CD16A.

Summary: Cancer stem cells (CSCs) are a small group of cancer cells with stem cell-like properties, which can be used for identifying malignant tumor phenotypes and patients with poor prognosis. Targeting CSCs has been shown to improve the effectiveness of cancer therapies. However, CSCs are resistant to conventional treatment methods such as radiotherapy and chemotherapy, hence the need for more effective anti-CSC therapies.

Summary: The overall survival of cancer patients depends on the success of therapy, with therapy failure linked to enhanced metastasis and recurrence. The innate immune system has been shown to play a key role in cancer cell communication and control of dormancy. New therapies targeting dormant cancer cells may hold promise for improving patient outcomes.

Summary: Pancreatic ductal adenocarcinoma (PDA) is a nearly universally lethal malignancy characterized by extensive infiltration of immunosuppressive myeloid cells, inhibiting normal immune functions and promoting carcinogenesis. Current immune checkpoint therapy has not been effective in PDA, highlighting the importance of understanding the mechanisms underlying immune suppression in this type of cancer.
