Microglia are the resident monocytic cells in the central nervous system (CNS), where they constitute the complex tissue structure together with a diverse set of cell types, including neurons, glial cells, and vasculature. Different from other cells, microglia have distinct features, including not only their origin but functions in the CNS, and serve multiple roles within healthy and diseased CNS tissue. The present review highlights the latest advance in our understanding of the nature of microglia in the CNS, from their origin to both the physiological and pathological roles throughout their whole life.

Summary: This study reveals the ontogenetic relationships and development of different subsets of macrophages in the central nervous system (CNS). Microglia and meningeal macrophages share a common prenatal progenitor, while perivascular macrophages originate from the perinatal meningeal macrophages after birth in an integrin-dependent manner. The presence of arterial vascular smooth muscle cells is crucial for the establishment of perivascular macrophages.

Summary: This study reveals the role of CD11c-expressing spinal microglia in the remission and recurrence of neuropathic pain. These microglia express IGF1, and interference with IGF1 signaling impairs pain recovery. Depletion of CD11c(+) microglia or interruption of IGF1 signaling leads to relapse in pain hypersensitivity.

Summary: The central nervous system (CNS) consists of neurons and various stromal cells, with microglia being a type of glial cells that play an important role in maintaining tissue homeostasis. They respond early to CNS abnormalities and can impact neuronal functions through their plasticity. There are also distinct macrophage populations at the border of the CNS, such as meninges and choroid plexus, which have different morphology and gene expression profiles compared to microglia. This review summarizes recent advances in understanding the heterogeneity, plasticity, and ontogenetic relationship of these CNS-resident macrophage populations.

Summary: The study reveals that diet-induced obesity impairs restoration of CNS demyelination by suppressing cholesterol efflux through TGF beta signaling, hindering microglial myelin-debris clearance and lesion recovery. Targeting TGF beta or promoting TREM2 activity could restore microglia responsiveness and improve myelin-debris clearance after demyelination.
