Compelling evidence suggests that benign prostatic hyperplasia (BPH) development involves accumulation of mesenchymal-like cells derived from the prostatic epithelium by epithelial-mesenchymal transition (EMT). Transforming growth factor (TGF)-beta induces EMT phenotypes with low E-cadherin and high vimentin expression in prostatic epithelial cells. Here we report that LPS/TLR4 signalling induces down-regulation of the bone morphogenic protein and activin membrane-bound inhibitor (BAMBI), which enhances TGF-beta signalling in the EMT process during prostatic hyperplasia. Additionally, we found that the mean TLR4 staining score was significantly higher in BPH tissues with inflammation compared with BPH tissues without inflammation (5.13 +/- 1.21 and 2.96 +/- 0.73, respectively; P < 0.001). Moreover, patients with inflammatory infiltrate were more likely to have a higher age (P = 0.020), BMI (P = 0.026), prostate volume (P = 0.024), total IPSS score (P = 0.009) and IPSS-S (P < 0.001). Pearson's correlation coefficient and multiple regression analyses demonstrated that TLR4 mRNA expression level was significantly positively associated with age, BMI, serum PSA levels, urgency and nocturia subscores of IPSS in the inflammatory group. These findings provide new insights into the TLR4-amplified EMT process and the association between TLR4 levels and storage LUTS, suggesting chronic inflammation as vital to the pathogenesis of BPH.

Comprehensive Profiling Reveals Prognostic and Immunogenic Characteristics of Necroptosis in Soft Tissue Sarcomas

Lin Qi, Ruiling Xu, Xiaolei Ren, Wenchao Zhang, Zhimin Yang, Chao Tu, Zhihong Li

Summary: This study identified necroptosis-related genes (NRGs) in soft tissue sarcomas (STSs) and found their potential role in prognosis. The results showed that the risk score based on the necroptosis-related gene signature could serve as an independent prognostic factor and could also predict immune infiltration. Furthermore, the necroptosis-related gene signature was validated in other malignancies, indicating its potential as a prognostic indicator in different types of cancer.

Summary: Denosumab, a monoclonal antibody used in the treatment of osteoporosis, has seen an increase in research publications over the past decade. The United States leads in denosumab research, with Amgen Inc. being the most influential entity. Clinical comparison and combination of denosumab with other drugs for osteoporosis treatment is a significant focus of research. Additionally, exploring the multiple roles of the RANK-RANKL-OPG system and optimizing drug administration strategies are potential future research directions.

Integrated Analysis of TME and Hypoxia Identifies a Classifier to Predict Prognosis and Therapeutic Biomarkers in Soft Tissue Sarcomas

Ruiling Xu, Lin Qi, Xiaolei Ren, Wenchao Zhang, Chenbei Li, Zhongyue Liu, Chao Tu, Zhihong Li

Summary: Soft tissue sarcomas are rare and aggressive cancers with high risks and poor prognosis. The tumor microenvironment (TME) and hypoxia have critical roles in tumor development. This study aimed to investigate whether linking hypoxia-related parameters to TME cells could improve prognosis and treatment outcomes. The Hypoxia-TME classifier was constructed and found to correctly classify patients based on prognosis and immune type. Moreover, specific immunotherapy and chemotherapy programs were provided. Several key genes were identified for further research. The Hypoxia-TME classifier demonstrated pretreatment predictive value for prognosis and therapeutic outcome, offering new approaches to therapy strategizing.

Chi Zhang, Xianfu Yi, Mengfan Hou, Qingyang Li, Xueying Li, Lu Lu, Enlin Qi, Mingxin Wu, Lin Qi, Huan Jian, Zhangyang Qi, Yigang Lv, Xiaohong Kong, Mingjun Bi, Shiqing Feng, Hengxing Zhou

Summary: By studying m(1)A RNA modification in normal and oxygen-glucose deprivation/reoxygenation (OGD/R) treated mouse neurons, we found that OGD/R increased the number of m(1)A-modified RNAs, and m(1)A modification may affect the regulatory mechanisms of noncoding RNAs and the translation of circRNAs. The systematic analysis of m(1)A landscape provides a critical foundation for understanding RNA modification and offers new perspectives and theoretical basis for treating and developing drugs for OGD/R pathology-related diseases.
