Tissue inflammation is a finely regulated process that controls wound healing and allows the clearance of damaged cells, pathogens and irritants. However, excessive or uncontrolled inflammation is detrimental, causing tissue damage and leading to autoimmunity. The recruitment of circulating leukocytes to the target tissue is a key stage in the inflammatory process, and is controlled by a multistep cascade in which adhesive receptors known as selectins mediate initial leukocyte tethering and rolling along vascular surfaces, which is required for their subsequent adhesion and arrest. This review considers the role of selectins and their ligands in the recruitment of circulating leukocytes to peripheral tissues during inflammatory responses that lead to the development of autoimmunity, focusing on data from animal models and clinical trials suggesting that selectins may offer valuable therapeutic targets for the treatment of autoimmune diseases. Metabolic determinants of leukocyte pathogenicity in neurological diseases

Marah C. Runtsch, Giovanni Ferrara, Stefano Angiari

Summary: Neuroinflammatory and neurodegenerative diseases involve the recruitment of circulating immune cells into the central nervous system, which can either perpetuate detrimental responses or dampen inflammation and promote tissue repair. Recent research highlights the role of metabolic reprogramming in regulating immune cell activity and function, with implications for potential therapeutic targeting in neurological disorders.

The Role of T Cell Senescence in Neurological Diseases and Its Regulation by Cellular Metabolism

Johannes Fessler, Stefano Angiari

Summary: Immunosenescence is a dysregulated state characterized by cell cycle arrest, telomere shortening, cellular stress markers expression, and pro-inflammatory mediators secretion. This process mainly occurs during aging, but can also be induced in other pathological conditions. Senescent immune cells may contribute to chronic inflammation and tissue damage, particularly in neurological diseases such as multiple sclerosis and Alzheimer's disease.

Summary: Itaconate and 4-octyl itaconate inhibit M2 macrophage polarization and metabolic remodeling by suppressing JAK1 phosphorylation and kinase activity. This discovery provides a new strategy to inhibit JAK1 in M2 macrophage-driven diseases.
