Characterization of Lysozyme-Like Effector TseP Reveals the Dependence of Type VI Secretion System (T6SS) Secretion on Effectors in Aeromonas dhakensis Strain SSU

Xiaoye Liang, Tong-Tong Pei, Zeng-Hang Wang, Weiliang Xiong, Li-Li Wu, Ping Xu, Shuangjun Lin, Tao G. Dong

Summary: The Type VI Secretion System (T6SS) is utilized by Gram-negative bacteria for interspecies interaction. This study identified a lysozyme-like effector TseP in Aeromonas dhakensis, showcasing its cell wall-lysing activities and T6SS dependency. Inactivating TseP and two other effectors led to the creation of a detoxified T6SS-active strain for potential protein delivery to various recipient cells.

Essential functions of chaperones and adaptors of protein secretion systems in Gram-negative bacteria

Kevin Manera, Fatima Kamal, Brianne Burkinshaw, Tao G. Dong

Summary: Protein secretion systems are vital for bacteria to interact with their environment through secreted toxic effectors. Accessory chaperone and adaptor proteins are essential for the proper folding and stability of effectors during the secretion process. Understanding the functions of these accessory proteins can lead to custom design of cargo effectors for biotechnological applications.

Sensing of intracellular Hcp levels controls T6SS expression in Vibrio cholerae

Kevin Manera, Florence Caro, Hao Li, Tong-Tong Pei, Steven J. Hersch, John J. Mekalanos, Tao G. Dong

Summary: The study reveals a quantity-sensing control mechanism of the T6SS, which down-regulates the expression of secreted components in the cytosol when T6SS is inactive. This Hcp-dependent sensing control allows organisms carrying T6SS to quickly adjust T6SS expression and prevent wasteful build-up of its major secreted components.

Engineered Type Six Secretion Systems Deliver Active Exogenous Effectors and Cre Recombinase

Steven J. Hersch, Linh Lam, Tao G. Dong

Summary: Genetic editing has been revolutionized by delivering Cre recombinase as a protein through engineered bacterial type six secretion system (T6SS). The versatile T6SS system can also deliver other exogenous effectors, like TseC, enhancing its efficacy against specific pathogens. This engineered system shows promise in microbiome manipulation and as a next-generation antimicrobial tool.

VgrG-dependent effectors and chaperones modulate the assembly of the type VI secretion system

Xiaoye Liang, Tong-Tong Pei, Hao Li, Hao-Yu Zheng, Han Luo, Yang Cui, Ming-Xuan Tang, Ya-Jie Zhao, Ping Xu, Tao Dong

Summary: The study on Aeromonas dhakensis strain SSU reveals that the specificity of effectors is dictated by the VgrG tail, and deletion of effector-specific chaperones severely impairs T6SS secretion. The research also shows that chimeric VgrG-effector pairs can bypass the requirement of heterologous VgrG complex and effector-stuffing inside the baseplate complex is crucial for T6SS assembly.

VgrG Spike Dictates PAAR Requirement for the Assembly of the Type VI Secretion System

Xiaoye Liang, Hao-Yu Zheng, Ya-Jie Zhao, Yi-Qiu Zhang, Tong-Tong Pei, Yang Cui, Ming-Xuan Tang, Ping Xu, Tao Dong

Summary: Multiple paralogs of the cone-shaped PAAR are found in gram-negative bacteria, but their necessity for T6SS assembly remains unclear. This study reveals a conditional requirement for PAAR in T6SS functions and suggests a core-auxiliary relationship among different PAAR-VgrG modules.

A Dueling-Competent Signal-Sensing Module Guides Precise Delivery of Cargo Proteins into Target Cells by Engineered Pseudomonas aeruginosa

Li-Li Wu, Shuangquan Yan, Tong-Tong Pei, Ming-Xuan Tang, Hao Li, Xiaoye Liang, Shuyang Sun, Tao Dong

Summary: We have developed a highly selective protein delivery platform, DUEC, which responds to direct contact of attacking cells by engineering the tit-for-tat/dueling response of H1-T6SS in Pseudomonas aeruginosa. By screening H1-T6SS-secreted substrates, we have created Tse6N as an effective secretion tag for delivering Cre. DUEC cells can selectively deliver the Tse6N-Cre cargo into T6SS+ but not T6SS- Vibrio cholerae cells, and can also deliver a nuclease cargo to selectively kill provoking cells in a mixed community.

Acquisition of T6SS Effector TseL Contributes to the Emerging of Novel Epidemic Strains of Pseudomonas aeruginosa

Anmin Ren, Minlu Jia, Jihong Liu, Tian Zhou, Liwen Wu, Tao Dong, Zhao Cai, Jiuxin Qu, Yang Liu, Liang Yang, Yingdan Zhang

Summary: This study characterized a P. aeruginosa clinical strain LYSZa7 and identified a T6SS lipase effector TseL(PA) that promotes bacterial pathogenesis by facilitating bacterial internalization into host cells. The findings suggest that clinical bacterial strains employ a diversified group of T6SS effectors for interbacterial competition and might contribute to the emergence of new epidemic clonal lineages.
