Summary: SAMBA has been identified as a plant-specific regulator of the anaphase-promoting complex/cyclosome (APC/C) in Arabidopsis and maize. Two samba genome edited mutants in maize showed growth defects, with samba-1 representing a knockout allele and samba-3 producing a truncated protein that still interacted with APC/C but with altered activity. This suggests a dosage-dependent role for SAMBA in controlling developmental processes.

Summary: This study reveals that a complex homeostatic balance between hematopoietic stem cells (HSCs) and hematopoietic progenitor cells is maintained through competition for a limited amount of cell signaling molecule. When early hematopoietic progenitors fail to interact with specific cells, HSC numbers increase.

Summary: Cell competition acts as a quality-control mechanism that eliminates cells less fit than their neighbors to optimize organ development. In the developing brain, endogenous cell competition occurs and correlates with the Axin2 expression level. Axin2 suppresses the p53 signaling pathway to maintain cell fitness, and its deficiency leads to cell elimination dependent on p53 signaling. The Axin2-p53 axis may coordinate cell fitness surveillance, regulate natural cell competition, and optimize brain size during neurodevelopment.

Summary: Invasive trophoblast cells play a crucial role in placental development and pregnancy outcomes. ASCL2 is identified as a key regulator of invasive trophoblast cell lineage development in humans and rats. ASCL2 deficiency impairs EVT cell differentiation and impacts placental morphogenesis.

Summary: Hematopoietic stem cells have the ability to self-renew or differentiate into various blood cell lineages. The cell fate decisions of self-renewal or differentiation must be tightly controlled to prevent diseases like bone marrow failure or leukemia. Studies have shown that the cell cycle length plays a crucial role in hematopoietic stem cell fate. Cell cycle kinetics, divisional history, asymmetric cell divisions, metabolic and organelle activity all have an impact on hematopoietic stem cell fate decisions.

Summary: Oligodendrocyte precursor cells (OPCs) migrate along blood vessels in the developing CNS, and the detachment of OPCs from vasculature is regulated by the formation of astrocytic endfeet on vessels, which allows OPC differentiation.

Summary: This study demonstrates the crucial role of Gata2 in the regulation of hematopoietic stem cell (HSC) development and function. Gata2 haploinsufficiency affects the transcriptional activation of HSCs and repression of endothelial cell identity, resulting in incomplete maturation of HSCs. Moreover, hematopoietic-specific induction of Gfi1b expression can restore HSC production in gata2b-null zebrafish embryos.

Summary: The self-renewal and differentiation of hematopoietic stem cells are tightly controlled by various cells and cytokines in the bone marrow microenvironment. Changes in the niche composition can lead to hematological malignancies, and processes like homing, proliferation, and differentiation of HSCs are crucial for the success of hematopoietic stem cell transplantation. Single-cell sequencing and in vivo imaging are powerful tools for studying the bone marrow microenvironment in diseases like leukemia and after transplantation.

Summary: This review article addresses the enigmatic role of the p53 transcription factor and tumor suppressor in programmed cell death during embryonic development. Contradictory results regarding the dependence of pluripotent embryonic stem cells on p53 for cell death are reconciled based on experimental conditions. The review concludes that p53 is not required for mouse embryonic development, but can accelerate programmed cell death in certain conditions. Additionally, DNA damage in differentiated cells induces p53-dependent cell cycle arrest and senescence.

Summary: Studies have revealed the role of Gpr56 in stem cell generation and differentiation, with its knockout leading to bias towards myeloid differentiation in mouse embryo HSCs, and an increased definitive myeloid progenitor cell frequency in Gpr56(KO) ESC cultures. Furthermore, mouse Gpr97 is able to rescue Gpr56 deficiency in hematopoietic generation in zebrafish.

Summary: A translocation at 9;11(p22;q23) leads to the formation of MLL-AF9 fusion protein, which is found in 25% of de novo AML cases in children. Using a hiPSC model with doxycycline-dependent MLL-AF9 expression, we investigated the effects of MLL-AF9 on hematopoietic development and its transformation to (pre-)leukemic states. We observed disruptions in early myelomonocytic development and identified gene profiles consistent with primary MLL-AF9 AML.
