Karyotype and AFLP data reveal the phylogenetic position of the Brazilian endemic Hypochaeris catharinensis (Asteraceae)

The genus Hypochaeris offers an excellent model for studies of recent adaptive radiation in the South American continent. We used karyotype analysis with chromomycin A(3) (CMA3)/4', 6-diamidino-2-phenylindole (DAPI) banding and fluorescence in situ hybridization (FISH), and amplified fragment length polymorphism (AFLP) fingerprinting to investigate for the first time the Brazilian endemic H. catharinensis and define its position within the South American group of species. Strong CMA-positive signals were seen at the end of both arms of chromosome 3 and at the end of the long arm of chromosome 4. DAPI bands were only detected in subterminal position on short arm of chromosome 4. FISH with 5S and 35S ribosomal DNA (rDNA) probes revealed a single 5S rDNA locus on short arm of chromosome 2, typical for all other South American Hypochaeris taxa analyzed to date. The 35S rDNA locus was identified at subterminal position on the short arm of chromosome 3, as reported so far for only two of the known species (H. lutea and H. patagonica). The AFLP study included 55 individuals, comprising nine species of the South American Hypochaeris plus their putative ancestor H. angustifolia. Eleven AFLP primer combinations generated a total of 401 fragments, of which 388 (96.7%) were polymorphic. High genetic similarities were observed among taxa, with all South American Hypochaeris species falling into one main cluster [100% bootstrap (BS)]. Hypochaeris catharinensis is closely related to H. lutea (82% BS), forming a well-separated subcluster within the South American species. Taken together, the karyological and AFLP data contribute to the placement of H. catharinensis within the phylogenetic framework of South American species of Hypochaeris and allow the definition of a novel and well-resolved phylogenetic group (the Lutea group).

Summary: This study investigated the ovule, seed, and fruit development in six Bulbostylis species, revealing variations mainly in pericarp development within the genus. Contrary to previous studies, the results suggest conflicts in infrageneric groupings based on fruit micromorphology and molecular analysis of the Bulbostylis species. This work enhances understanding of reproductive anatomy and embryology in Bulbostylis and sheds light on the origin of multiple embryos in Cyperaceae.

Molecular and Cytogenetic Analysis of rDNA Evolution in Crepis Sensu Lato

Summary: This study investigated the chromosome evolution of Crepis plants by analyzing the chromosome base number and organization of rDNA loci in 39 species. The results showed that despite variations in rDNA loci organization, most species had rDNA loci on the same chromosome arm. Comparative analysis revealed multiple independent gains of rDNA loci and repositioning during diversification and speciation. These findings provide important insights into the evolutionary mechanisms and speciation of Crepis plants.

Repeat Dynamics across Timescales: A Perspective from Sibling Allotetraploid Marsh Orchids (Dactylorhiza majalis s.l.)

Summary: In this study, the fate of transposable elements (TEs) in Dactylorhiza allotetraploids formed through hybridization between diploid species D. fuchsii and D. incarnata was investigated. The results showed that the paternal genome had a higher content of LTR retrotransposons compared to the maternal genome, resulting in a larger genome size. The genome size and TE composition in the youngest allopolyploid appeared to be additive, while rampant genome expansion was observed in intermediate-aged polyploids. A non-autonomous tandem repeat element played a major role in the genome dynamics of the allopolyploids. Overall, the fate of repeats in this system was strongly regulated and predictable across multiple independent allopolyploidization events. Mild genomic shock was also observed following allopolyploidization, possibly due to strong selection against further genome expansions.

Comparative Analyses of Complete Chloroplast Genomes and Karyotypes of Allotetraploid Iris koreana and Its Putative Diploid Parental Species (Iris Series Chinenses, Iridaceae)

The Pyrenees as a cradle of plant diversity: phylogeny, phylogeography and niche modeling of Saxifraga longifolia

Summary: In this study, we investigated the colonization history of Saxifraga longifolia, a plant species abundant in the Pyrenees and rare in other Iberian mountains and the African Atlas. Our findings suggest that the current distribution is the result of the shrinkage of a more extensive distribution in previous cold periods. We also discovered seven plastid haplotypes, predominantly found in the Pyrenees, and inferred migration routes mainly from the Pyrenees to other areas.

Phenolic compounds and color of labeled resin spurge honey and their correlations with pollen content

Diversification of chiles (Capsicum, Solanaceae) through time and space: New insights from genome-wide RAD-seq data

Summary: This study presents a new and updated phylogenetic hypothesis on the evolution of the Capsicum genus, incorporating nearly all currently accepted species. The study proposes a new model of spatial diversification based on genome-wide analysis. The results suggest that the Northern and Central Andes were the likely ancestral range of Capsicum, and that the diversification of the genus began in the Pliocene and continued through the Pleistocene.

Chromosomal and genomic data in Neotropical Dorstenia species (Moraceae) and their relationships with African species

Summary: This study investigated the karyotype diversity and DNA C values of Dorstenia spp. from the Neotropical region and examined the genomic sequencing of African and Neotropical species. The results showed that polyploidy plays a significant role in karyotype differentiation, but there is no clear relationship between DNA C values and karyotype features or rDNA sites. Dysploidy events and rearrangements involving repetitive fractions are suggested to be responsible for chromosome number variations and changes in genome size.

Genome-environment associations along elevation gradients in two snowbed species of the North-Eastern Calcareous Alps

Summary: This study examines the genetic structure, diversity, and genome-environment associations of two snowbed species in the Eastern Alps, and finds that they are suitable models for genetic monitoring of climate change adaptation along elevation gradients. The main effects of climate change on these species are changes in precipitation and sunlight exposure, leading to alterations in snow cover duration and encroachment of shrubs. Further research is needed to characterize and validate the genomic loci involved in adaptive processes by assembling genomes and studying larger sample sizes and time series.

Evolutionary features of microspore and pollen grain development in Cyperaceae

Summary: Asymmetric meiosis leading to the release of pseudomonad pollen grains is a shared feature in Cyperaceae. This study analyzed the early- and late-diverging sedge genera Hypolytrum and Eleocharis, respectively, and compared them to other genera in the family. The results show variations in pseudomonad shape and positioning, with homologous development in Hypolytrum and unique traits in Eleocharis, but shared ultrastructural features among the genera.
