Prostate cancer is the most diagnosed noncutaneous cancer among men in Western developed countries. Nutrition and environmental factors play a major role in the onset of the disease, but the molecular details for the contribution of each factor is elusive. In an effort to better understand the basic biology of prostate cancer, we identified a new protein family that is named the 6 trans-membrane protein of prostate (STAMP) that appear to have important functions in prostate cancer. At least one member of the STAMP family is also implicated in metabolic homeostasis and nutrition response. Here, we review the current biology of the STAMP proteins and how they may be implicated in disease states including cancer and metabolic syndrome.

Summary: Prostate cancer is one of the most common non-skin cancers among men, with no curative treatment currently available. STAMP2, an important factor for prostate cancer growth and survival, is regulated by inflammatory signaling which may affect disease progression. Inflammatory events and dysregulated cytokine expression are implicated in prostate cancer, and STAMP2 knockdown increases the sensitivity of prostate cancer cells to cytokine treatment.

Summary: In prostate cancer cells, activating transcription factor 4 (ATF4) exclusively activates gene expression involved in the mitochondrial (m1C) one-carbon cycle, not the cytoplasmic (c1C) cycle; knockdown of methylenetetrahydrofolate dehydrogenase 2 (MTHFD2) inhibits prostate cancer cell growth significantly. This suggests that the m1C cycle plays a key role in prostate cancer progression and may serve as a potential biomarker and therapeutic target.

Anticarin-β shows a promising anti-osteosarcoma effect by specifically inhibiting CCT4 to impair proteostasis

Summary: Unlike healthy cells, cancer cells heavily rely on the molecular chaperone protein TRiC, particularly its subunit CCT4, for protein folding and maintenance of proteostasis. In this study, we demonstrate that targeting CCT4 with the compound anticarin-beta can effectively inhibit osteosarcoma cell survival and impede STAT3 maturation. Anticarin-beta shows high selectivity for cancer cells and exhibits remarkable antitumor efficacy in orthotopic and patient-derived xenograft models of osteosarcoma.

Clinical significance of EPHX2 deregulation in prostate cancer

Summary: The EPHX2 gene, frequently deleted in prostate cancer, shows significantly decreased expression levels in tumors compared to normal tissues, and is inversely correlated with Gleason grade and disease-free survival time. Its deregulation is significantly correlated with the clinical characteristics of PCa progression and may serve as a prognostic marker for PCa.
