Summary: The combination of mechanical fluid shear stress with biomaterials promoted osteogenic differentiation of MSCs, leading to increased cell viability and migration. Osteogenic induction with biomaterials under mechanical fluid shear stress showed enhanced Runx2 protein expression, suggesting a potential clinical application for bone defect treatment. The study demonstrated that different biomaterial scaffolds influenced MSC morphology, viability, and osteogenic differentiation markers in varying ways under different culture conditions.

Summary: We developed a highly efficient platelet lysate-based approach to directly transdifferentiate skin-derived human fibroblasts to osteoblast-like cells. The transdifferentiated cells showed increased expression of osteoblast-specific markers and exhibited osteogenic properties. RNA-seq analysis revealed distinct transcriptome profiles between the transdifferentiated cells and primary fibroblasts. This in vitro model may contribute to studying osteoblast-dependent disorders in patient-derived cells.

Summary: In this study, a novel human mesenchymal stem cell (hMSC)-laden GO/alginate/gelatin composite bioink was developed for forming 3D bone mimicking scaffolds. The GO composite bioinks showed improved bioprintability, scaffold fidelity, cell proliferation, osteogenic differentiation, and ECM mineralization. The optimal GO group was 1GO, which demonstrated great potential for 3D bioprinting of bone tissue models and tissue engineering applications.

Summary: Tunable mechanical properties of hydrogels play a key role in regulating stem cell differentiation. By modulating cellular spatial sensing of integrin-adhesive ligands on low stiffness hydrogels, the mechanosensing and osteogenic differentiation of MSCs can be promoted to levels comparable with those observed on stiffer hydrogels. This approach opens up new possibilities for designing biomaterials and tissue scaffolds for regenerative therapeutics.

Summary: Developing a silk fibroin-based coating embedded with nanohydroxyapatite and silver nanoparticles is an effective approach to enhance bone healing and inhibit bacterial infection. The coating showed good antibacterial ability by inhibiting bacterial adhesion and killing planktonic bacteria efficiently. Additionally, it was beneficial for osteoblast adhesion, spreading, and proliferation, while reducing the cytotoxicity of silver nanoparticles.
