Derricin and Derricidin Inhibit Wnt/β-Catenin Signaling and Suppress Colon Cancer Cell Growth In Vitro

Overactivation of the Wnt/beta-catenin pathway in adult tissues has been implicated in many diseases, such as colorectal cancer. Finding chemical substances that can prevent this phenomenon is an emerging problem. Recently, several natural compounds have been described as Wnt/beta-catenin inhibitors and might be promising agents for the control of carcinogenesis. Here, we describe two natural substances, derricin and derricidin, belonging to the chalcone subclass, that show potent transcriptional inhibition of the Wnt/beta-catenin pathway. Both chalcones are able to affect the cell distribution of beta-catenin, and inhibit Wnt-specific reporter activity in HCT116 cells and in Xenopus embryos. Derricin and derricidin also strongly inhibited canonical Wnt activity in vitro, and rescued the Wnt-induced double axis phenotype in Xenopus embryos. As a consequence of Wnt/beta-catenin inhibition, derricin and derricidin treatments reduce cell viability and lead to cell cycle arrest in colorectal cancer cell lines. Taken together, our results strongly support these chalcones as novel negative modulators of the Wnt/beta-catenin pathway and colon cancer cell growth in vitro.

Therapeutic Potential of Naturally Occurring Small Molecules to Target the Wnt/beta-Catenin Signaling Pathway in Colorectal Cancer

Summary: Colorectal cancer (CRC) is the second leading cause of death worldwide, and its treatment is limited due to late diagnoses and resistance to available treatments. The use of naturally occurring small molecules that inhibit the Wnt/beta-catenin pathway in experimental CRC models has shown promise in overcoming these challenges. However, moving these small molecules from the laboratory to the clinical platform presents its own set of progress and challenges. This review discusses the importance of understanding the role of the Wnt/beta-catenin pathway in CRC and highlights the potential of naturally occurring small molecules as therapeutic agents.

Summary: The ethanol extract from Euphorbia tirucalli L. and its latex suspended in water are used in Brazil for the treatment of various diseases, including cancer. This study aimed to determine the cytotoxic effects of the ethanol extract on gastric adenocarcinoma cells (AGS) and analyze its chemical composition. Hexane and aqueous fractions of the extract showed higher cytotoxicity on AGS cells. Analysis revealed the presence of triterpene compounds in the hexane fraction and ellagic acid as the main phenolic compound in the aqueous extract, suggesting that these components contribute to the greater cytotoxic activity of the ethanol extract compared to latex.

Summary: This study evaluated the quality of Brazilian peppertree fruits at different maturation stages and found that the red fruits had the highest yield of essential oil. The analysis of physical and chemical characteristics of each stage showed distinct profiles, with the pinkish-green and pinkish stages displaying a transition in characteristics. The chromatography data were suitable for chemometric analysis and can contribute to optimizing the harvest time for aroeira fruits.
