Summary: Atherosclerosis is a leading cause of death globally, and there is a correlation between periodontal disease and atherosclerosis. This study used Porphyromonas gingivalis-derived LPS (Pg-LPS) to investigate the proliferation and migration of human aortic smooth muscle cells (HASMCs). The results suggest that Pg-LPS promotes atherosclerosis through the activation of the TLR4-MAPK signaling pathway.

Summary: The local environment surrounding airway smooth muscle (ASM) cells plays a significant role in influencing the physiological and phenotypic properties of the ASM tissues. The cells have the ability to adapt and modify their properties in response to the mechanical forces and factors present in their extracellular milieu. Membrane adhesion junctions between smooth muscle cells and the extracellular matrix (ECM) not only provide mechanical coupling but also sense and transmit environmental signals to intracellular signaling pathways.

Summary: Vessel wall calcification is a structural pathology of aged vessels associated with high cardiovascular mortality in elderly patients. Chronic inflammation and oxidative stress in aging lead to activation of PRRs in vascular cells, contributing to VSMC calcification. Inhibition of PRR activation may be a potential target for reducing vascular calcification and subsequent cardiovascular mortality.

Summary: DOT1L inhibition in VSMCs significantly reduces atherosclerosis progression by directly modulating Nf-kappa B1 and Nf-kappa B2 transcription, which are master regulators of inflammation inducing expression of CCL5 and CXCL10 cytokines, key in atherosclerosis development. DOT1L could be a promising therapeutic target for vascular diseases as its inhibition reduces plaque progression.

Summary: Glutamate receptors and related enzymes were identified in VSMCs from human atherosclerotic lesions, with AMPA-type glutamate receptors playing a role in VSMC phenotypic modulation. Lower mRNA levels of GRIA1 and GRIA2 in atherosclerotic lesions were associated with adverse clinical events, suggesting the importance of further exploring neurotransmitter signaling in the pathogenesis of human atherosclerosis.

Summary: Considerable evidence suggests that cognitive impairment is primarily a vascular disorder. The depletion of SM22 alpha in VSMCs contributes to their phenotypic switching and inflammation, but the role of VSMCs in cognitive impairment is still uncertain. This study shows a possible link between VSMC phenotypic switching and neurodegenerative diseases, supporting the idea of VSMCs and SM22 alpha as therapeutic targets in cognitive impairment.

Additionally, the natural inhibitor of TMEM16A, arctigenin, can lower blood pressure and alleviate vascular inflammation in salt-sensitive hypertension mice.
