Summary: Deletions of chromosome 17p, especially in TP53 gene, are frequently observed in human cancers and associated with poor outcomes. Previous studies have shown that p53-independent mechanisms are involved in 17p deletions-driven cancers. In this study, we found that altered arachidonate metabolism caused by Alox8 deficiency on chromosome 11B3 contributes to B cell malignancy. Upregulation of the cyclooxygenase pathway and increased levels of prostaglandin E2 were observed in cells with Alox8 loss. Ectopic PGE2 inhibited apoptosis and differentiation of pre-B cells. The upregulation of Cox-2 by Alox8 deficiency was found to be crucial for tumorigenesis, and COX-2 inhibitor was effective in treating tumors with Alox8 or 11B3 loss.

Summary: Tuberculosis caused by Mycobacterium tuberculosis is a global health concern. In this study, it is shown that M. tuberculosis induces COX-2 expression in human macrophages and promotes the formation of anti-inflammatory J2-PGs. These J2-PGs have anti-inflammatory effects in M1-MDMs, downregulating proinflammatory cytokines and upregulating anti-inflammatory cytokines and M2 markers.

Summary: This study found that elevated plasma levels of PGE(2) are expressed in half of patients with OPC secondary to overexpression of COX-2 by peripheral blood monocytes, and this expression is driven by IL-1 alpha secreted by the tumors. Monocytes from patients are much more sensitive to the stimulation than monocytes from controls, suggesting the possibility of enhanced immune-modulating feed-back loops. Furthermore, control monocytes pre-exposed to PGE(2) overexpress COX-2 in response to IL-1 alpha, simulating responses made by monocytes from some OPC patients.

Summary: This study investigated the potential of plant-derived components ZBN, ELA, and QCT to modulate COX-3 and cytokines associated with arthritic disorders. In-vitro models and in-silico docking studies supported the anti-inflammatory effects of these molecules. In-vivo experiments confirmed the anti-arthritic potential of ZBN and ELA in arthritic rats, with significant reductions in TNF-a and IL-1 beta levels observed. ELA also attenuated COX-3 upregulation, while ZBN downregulated NF?B expression in arthritic animals.
